Mesangial cells (MCs) are essential for maintaining glomerular homeostasis and play a critical role in initiation and progression of glomerular disease. However, it remains unclear how the MCs are regulated under pathologic conditions. Here, we show that selective hyperactivation of YAP/TAZ in PDGFRβ+ MCs results in dedifferentiation into myofibroblast, which leads to mesangial cell proliferation and mesangial fibrosis. In addition, mesangial expansion induces impairment of glomerular vascular barriers and loss of glomerular filtration function. Furthermore, transcriptomic and biochemical analyses reveal that YAP/TAZ and N-MYC interaction is required for development of mesangial expansion. YAP/TAZ adjust the half-life of N-MYC to cause MC proliferation and following fibrosis. Together, our findings demonstrate the molecular significance of YAP/TAZ and N-MYC in mesangial cell dedifferentiation and fibrosis, providing the insight to describe regulatory mechanisms under glomerular sclerosis.
사구체에 존재하는 혈관사이세포는 사구체 족세포 및 사구체 혈관내피세포와 활발하게 상호작용하며 사구체의 생태를 조절하고 있다. 당뇨성 신증에서는 혈관사이세포가 과도하게 증식하여 사구체 구조적 변화와 함께 기능적 저하를 유발하는 것으로 알려져 있지만, 병태생리와 관련된 기전은 아직 잘 밝혀져 있지 않다. 본 연구에서는 사구체 내 혈관사이세포가 증식하는데 기여하는 분자적 기전을 밝히고자 하였다. Hippo pathway는 세포 증식과 근섬유아세포의 분화, 조직 섬유화 등에 관련되어 있으며 YAP/TAZ는 Hippo pathway의 주요 하위 신호 전달자이다. 신장 섬유화가 발생하는데 있어 YAP/TAZ 가 수행하는 역할은 일부 규명되어 있으나 사구체 내 혈관사이세포에서 YAP/TAZ의 역할은 아직 밝혀지지 않았다. 본 연구에서는 당뇨성 신증이 진단된 사구체에서 YAP 발현이 증가되어 있는 것을 확인하였으며, YAP/TAZ를 과발현 시킨 유전자 조작 실험동물을 통해 당뇨병성 신증과 유사한 질병 상태를 모사하였다. 또한 전사체 분석 및 생화학적 분석을 통하여 YAP/TAZ의 하부 신호전달에 N-MYC 이 중요한 역할을 하고 있음을 밝혀내었다. 이 연구에서 가장 중요한 것은 YAP/TAZ와 N-MYC 이 상호작용하여 사구체 내 혈관사이세포 증식이 발생하며, 2차적으로 혈관내피세포와 족세포에 변형을 초래한다는 것을 발견하여, 추후 혈관사이세포 증식을 조절할 수 있는 유망한 분자기전을 밝혀내었다는 점이다.
