Optogenetic activation of receptors has advantages compared to chemical or ligand treatment with high spatial and temporal precision. Especially in the brain, using genetically expressed light tunable receptors is superior to direct infusion or systemic drug treatment.
In chapter 1, we applied light activatable TrkB receptor, Opto-cytTrkB(E281A), in mouse brain with reducing basal activity by recruiting Cry2PHR(E281A) mutant and deleting extracellular domain to prevent it from unexpected activation by endogenous ligand. This form was sufficiently expressed in the mouse hippocampus and the cortex via viral gene delivery with highly sensitive toward blue light even non-invasive light stimulation fulfilling canonical signal transduction. Also, we demonstrated Opto-cytTrkB(E281A) can locally induce TrkB signaling in axon terminals in the MEC-HPC circuit. From these results, we suggest that Opto-cytTrkB(E281A) will be useful to elucidate the time- and region-specific role of TrkB signaling from cellular function to neural circuit mechanisms.
It takes a long-time for the memory to be remote memory. The frontal cortex is thought to be the region where the old memory is recalled. However, the TrkB underlie the neural circuit remains to be elucidated.
In chapter 2, we suggest that TrkB in neurons projecting the entorhinal cortex to prefrontal cortex is required to consolidate remote memory by increasing reactivity of mPFC cells. Furthermore, TrkB deletion devastated the experience-dependent maturation of oligodendroglia in the prefrontal cortex resulting in defect of remote memory, which was restored by clemastine administration. Together, our data suggest TrkB in the intercortical circuits served the remote memory consolidation.
수용체를 빛 활성화시키는 것은 뇌 연구에 있어서, 빛을 이용한 방법은 직접 주사 혹은 체순환 투여보다 매우 효과적이다. 첫 번째 챕터에서는 빛으로 활성 가능한 TrkB 수용체인 Opto-cytTrkB(E281A)를 단백질 변형체 적용과 세포 외 도메인을 제거 후 바이러스 벡터를 이용한 발현 시 생쥐 뇌에서 부작용 없이 작동함을 확인하였다. 이 형태의 발현은 빛에 매우 민감하였다. 또한 내후각피질-해마 신경회로 뉴런의 신경말단 부위를 빛으로 자극하여 수용체의 신호전달 기작을 유도할 수 있었다. 따라서 Opto-cytTrkB(E281A)는 신경회로 메커니즘 규명에 있어 시공간적으로 신호전달을 조절할 수 있다는 점에서 장점이 있음을 제시한다.
전두엽 피질의 장기기억 저장 역할에 대한 정확한 분자적 메커니즘은 아직 잘 밝혀져 있지 않다. 두 번째 챕터에서는 TrkB 수용체의 장기기억 강화 메커니즘을 제시한다. 이 수용체를 피질 간 신경회로에서 제거 시, 전두엽 피질에서의 신경세포의 재활성화가 줄어들었다. 또한 희소돌기아교세포의 생성과 성장이 줄어들었음을 확인하였다. 따라서 이 수용체의 뇌 피질 간 신경회로에서의 장기 기억 강화 메커니즘에 대해 제시한다.
