Basal-like breast cancer is the most aggressive subtype with the worst prognosis. Despite its high recurrence rate, chemotherapy is the only treatment for basal-like breast cancer, which lacks expression of hormone receptors. In contrast, luminal-A tumors express ERα and can undergo endocrine therapy for treatment. Previous studies have tried to develop effective treatments for basal-like patients using various therapeutics but failed due to the complex and dynamic nature of the disease. In this study, we performed a transcriptomic analysis of breast cancer patients to construct a simplified but essential molecular regulatory network model. Network control analysis identified potential targets and elucidated the underlying mechanisms of reprogramming basal-like cancer cells into luminal-A cells. Inhibition of BCL11A and HDAC1/2 effectively drove basal-like cells to transition to luminal-A cells and increased ERα expression, leading to increased tamoxifen sensitivity. High expression of BCL11A and HDAC1/2 correlated with poor prognosis in breast cancer patients. These findings identify mechanisms regulating breast cancer phenotypes and suggest the potential to reprogram basal-like breast cancer cells to enhance their targetability.
Basal-like 유방암은 유방암 아형 중 가장 악성이고 예후가 안좋은 것으로 알려져 있다. 또한 이는 높은 재발률을 가지고있지만 호르몬 수용체의 발현이 없어서 화학요법 외에는 다른 치료법이 없다고 알려져 있다. 반대로, luminal-A 아형으로 가지는 유방암은 ERα 호르몬 수용체의 발현으로 이를 타겟하는 호르몬 요법을 통해 치료 받을 수 있다. 기존 연구들은 다양한 치료법을 사용하여 basal-like 환자에 대한 효과적인 치료법을 개발하려고 시도했지만 아직 다 밝혀지지 않은 이 질병의 복잡하고 역동적인 유전자와 분자의 관계적 특성으로 인해 실패했다. 이 연구에서 우리는 단순하지만 필수적인 분자 조절 네트워크 모델을 구축하기 위해 유방암 환자의 전사체 분석을 수행했다. 네트워크 제어 분석은 잠재적인 표적을 식별하고 basal-like 암세포를 luminal-A 세포로 리프로그래밍하는 기본 메커니즘을 설명했다. BCL11A 및 HDAC1/2의 억제는 효과적으로 basal-like 세포를 luminal-A 세포로 전환하고 ERα 발현을 증가시켜 타목시펜 약물의 민감성을 증가시켰다. BCL11A 및 HDAC1/2의 높은 발현은 유방암 환자의 나쁜 예후와 상관관계가 있었다. 이러한 발견은 유방암 표현형을 조절하는 메커니즘을 확인하고 basal-like 유방암 세포를 리프로그래밍하여 표적화 가능성을 높일 수 있음을 시사한다.
