Bone marrow (BM) is commonly utilized in the clinical situation in the form of bone marrow transplantation (BMT) to treat patients with various hematological disorders. Unfortunately, a number of post-transplant complications mainly occur due to lack of initial engraftment at early post-transplant phase, which has been a critical limiting factor for achieving successful BMT outcomes. However, underlying cellular engraftment dynamics which be the beginning point to alleviate several complications and enhance hematopoietic recov-ery have not been fully identified yet. In this research, we performed a longitudinal in vivo imaging of spatio-temporal cellular behaviors of transplanted BM cells during early engraftment by utilizing a custom-built real-time laser-scanning confocal microscopy system.
We successfully performed a longitudinal nuclear-cytoplasmic visualization of transplanted BM cells with a H2B-GFP/β-actin-DsRed double-transgenic mouse model and fluorescently labeled vasculature landmark. Longitudinal in vivo BM monitoring clearly delineated the dynamic cellular behaviors of transplant BM dur-ing engraftment including cluster formation, migration, active proliferation, and differentiation into certain cell types in vivo.
We visualized multi-phase spatiotemporal cellular dynamics of transplanted hematopoietic stem and progen-itor cells (HSPCs) during early engraftment. A daily longitudinal wide-area intravital imaging of host cranial BM until post-transplant day 10 identified two distinctive behaviors of transplanted HSPC groups including cluster formation and cluster dissociation in vivo. Especially, in vivo cellular dynamics of cluster formation followed by cluster dissociation and serial de novo cluster formation around the dissociation spot were ob-served with coordinated vascular dynamics. Finally, we performed a multi-position long-term time-lapse in vivo visualization of transplanted HSPCs on transplant day 2-4 when the cluster formation occurred, which revealed three collective spatiotemporal cellular phases of initial engraftment with distinct cellular density positively coordinated with proliferation rate and negatively correlated with migration velocity.
골수는 임상에서 다양한 혈액 질환을 가진 환자들의 치료에 골수 이식의 형태로 널리 이용되어 왔다. 불행히도, 수 많은 이식 합병증은 주로 이식 초기 단계에 초기 생착의 불완전함에 의해 일어나고, 이는 성공적인 이식 결과 달성에 치명적인 제한 요소로 작용해왔다. 하지만, 여러 이식 합병증을 완화하고 조혈 회복 향상의 기반이 되는 이식 초기 생착 과정에 대한 이해는 온전히 밝혀지지 않았다. 본 연구에서는 자체 제작한 실시간 레이저 주사 공초점 현미경을 이용하여 생체 내 이식골수세포의 시공간적 생착 양상 영상화를 진행하였다. H2B-GFP/β-actin-DsRed 마우스 모델과 혈관 표지를 이용하여 시간에 따른 이식골수세포의 핵-세포성 영상화를 진행하였다. 그 결과, 생착 과정에서 이식 골수세포의 세포 군집 형성, 이동 양상, 활발한 분화 및 증식의 동적 양상을 관찰하였다. 또한, 이식 조혈 줄기 및 전구 세포의 단계별 초기 생착의 동적 양상을 시공간 분해능으로 관찰하였다. 이식 10 일차에 걸친 이식 수혜자 두개골 골수의 광역 이미징을 통해 생체 내 조혈 줄기 전구 세포의 두가지 대표적인 행동 양상인 세포 군집 형성 및 분열 과정을 확인하였다. 특히, 세포 군집 형성 후 급격한 군집 분열의 양상과 분열 지점 주변으로 새로운 여러 군집 형성의 양상을 관찰하였고, 이는 혈관 변화 양상과 동시에 일어남을 확인하였다. 마지막으로 세포 군집 형성이 시작되는 이식 2-4 일차에 이식 조혈 줄기 전구 세포의 장기간 다중 위치 생체 내 영상화를 진행하여 3가지 초기 생착 단계에 대한 시공간 세포 양상을 확인하였고, 이식 세포의 위치적 특성에 따른 증식률의 증가와 세포성 이동률 감소의 연관관계를 입증하였다.
