Colorectal cancer (CRC) is one of the most extensively studied model for staging genetic mutations along cancer progression. Yet, how CRC initiates still remains elusive because of its innate nature of rarity and difficulties in observation. To understand the earliest events in CRC initiation, we established early CRC development model by downregulating adenomatous polyposis coli (APC) in normal human colorectal epithelial, and performed single cell RNA sequencing (scRNA-seq). Interestingly, two subpopulations with different growth rate were found in single cell data: fast growth subpopulation (FG) and slow growth subpopulation (SG). These two subpopulations were characterized using scRNA-seq analysis tools and systems biology tools, and it turned out that FG is responsible in cancer development advance whereas SG eventually perish. Therefore, we conducted master regulator analysis to find candidate genes which are able to drive FG into SG, and then validated them with in vitro experiment. This study provides not only an insight on the early CRC development but also new therapeutic strategy which restrains cancer to advance further.
대장암은 발달 과정이 비교적 잘 알려진 고형암 중 하나이다. 그럼에도 대장암 발생 시점의 사건들은 그 희귀성과 관찰의 어려움 때문에 아직 많은 부분들이 분명하지 않다. 본 연구에서는 대장암 발생 초기 단계에서 벌어지는 사건들을 포착하기 위해 정상 대장 세포에서 adenomatous polyposis coli(APC)가 결실된 대장암 발달 초기 모델을 구축하고, 단일 세포 RNA sequencing 기술을 이용하여 대장암 발생 초기의 변화를 추적하였다. 그 결과 APC 결실 이후 단기간 내에 빠른 성장 세포군과 느린 성장 세포군이 나타나는 것을 발견하였다. 단일 세포 전사체 분석 기법과 시스템 생물학 분석을 이용하여 두 세포군의 특징을 분석하여, 빠른 성장 세포군이 암 발달 심화에 기여하는 반면 느린 성장 세포군은 더 발달하지 못하고 도태된다는 것을 밝혔다. 이에 빠른 성장 세포군을 느린 성장 세포군으로 전환하는 목표를 수립하고, 둘 사이에서 차이를 보이는 핵심 조절자를 탐색하여 세포 실험을 통해 그 실제 효과를 검증하였다. 본 연구는 대장암 발생 초기 단계에 대한 통찰을 제공함과 동시에 암을 비증식 상태로 강제하는 새로운 치료 전략을 제시한다.
