Fluorescent molecules have fluorescence and photothermal properties depending on the emission pathway of the absorbed energy. Such fluorescence and photothermal characteristics of fluorescent moles are widely used in the field of bio-imaging and photothermal treatment, respectively. In this thesis, research on fluorescence imaging and hydrogel actuation using multiplexing of these fluorescence and photothermal properties is demonstrated. First, a study on simultaneous multiplexed signal amplification for ultra-high-resolution 3D molecular imaging is shown. Linear amplification of three fluorescence signal intensities is induced by cyclic staining of the secondary antibodies labeled with a fluorescent molecule. Second, a study on selective control of hydrogel actuator by three different wavelengths of light is demonstrated. Actuation is induced by volumetric shrinkage of temperature-responsive hydrogels using wavelength-selective photothermal effect of three fluorescent molecules.
형광분자는 인가된 에너지를 흡수한 후 방출하는 경로에 따라 형광 특성 및 발열 특성을 갖는다. 이러한 형광 특성은 바이오 이미징 분야에서, 발열 특성은 광열 치료에 널리 사용되고 있다. 본 학위 논문에서는, 이러한 형광 및 광열 특성의 다중화(multiplexing)을 이용한 형광 이미징 및 하이드로젤 액츄에이션에 대한 연구에 대해 다룬다. 첫번째로, 초고해상도 3D 분자 이미징을 위한 동시 다중 신호 증폭 기술에 대한 연구를 보여준다. 형광분자가 표지된 2차 항체를 반복적으로 염색함으로써 세 가지 형광 신호 세기의 선형적인 증폭을 유도한다. 두번째로, 세 파장대의 빛에 의해 선택적으로 조절되는 하이드로젤 액츄에이션에 대한 연구를 보여준다. 세 가지 형광분자의 파장 선택적 광열효과를 이용해 온도 감응성 하이드로젤의 부피 수축을 통한 액츄에이션을 유도한다.
