The biophysical properties of the tumor microenvironment (TME) have been increasingly focused on due to their potential impacts on cancer progression. Biophysical cues in the TME such as stiffness, extracellular matrix (ECM) structure, and solid stress not only play a role in the regulation of cancer cells but also modulate the formation and activation of cancer-associated fibroblasts (CAFs). However, the effect of the physical constraint (PC) of the matrix on the activation of CAFs remains ambiguous. In this study, we investigated the transformation of normal fibroblasts into a CAF-like phenotype under the stimulation of cancer secretome with or without PC. We found that in the PC condition, stimulated fibroblasts caused local ECM disruption and reduced matrix stiffness, leading to less activation of the mechanical signaling pathway. Meanwhile, without PC, fibroblasts were able to induce stiffened matrix and thus much more mechanically active. Interestingly, this PC-free condition also induced a remarkably high expression profile of ECM-remodeling genes in fibroblasts. Our findings indicate the importance of PC in the induction of different CAF phenotypes and suggest the necessary consideration of PC in building models for CAFs studying.
종양 미세환경의 생물물리학적 특성은 암 진행에 중요한 역할을 하는 것으로 알려져 있다. 종양 미세환경의 강성, 세포외 기질 구조와 같은 생물물리학적 특성은 암세포뿐만 아니라 암 관련 섬유아세포 형성과 활성화에도 영향을 미친다. 하지만 암 관련 섬유아세포의 활성화와 세포외 기질의 물리적 제약에 대한 효과는 여전히 알려진 바가 많지 않다. 이 연구에서는 물리적 제약의 여부와 관계없이 암 분비체 자극 하에 정상 섬유아세포가 암과 유사한 표현형으로 변하는 것을 확인했다. 물리적 제약을 가한 조건에서는 섬유아세포가 세포외 기질 단백질을 국소적으로 붕괴시키는 것을 관찰하였고, 배양 매트릭스의 전체적인 강성이 낮아지며 기계적 신호전달 경로의 활성화도 감소하는 것을 확인했다. 그에 반해 물리적 제약이 없는 조건의 섬유아세포는 주변 매트릭스를 더욱 단단하게 만들었고, 기계적 신호전달 경로도 더 활성화되는 것을 관찰했다. 흥미롭게도, 물리적 제약이 없는 조건에서 섬유아세포의 세포외 기질 리모델링 관련 유전자 발현이 더 높게 나타났다. 이 연구 결과는 다양한 암 관련 섬유아세포 표현형 유도에 물리적 제약이 중요한 조건임을 나타내고 있고, 앞으로의 관련 연구에서도 물리적 제약 조건을 고려해야 더 생체적으로 유사한 플랫폼을 구축할 수 있을 것으로 생각된다.
