Extracellular matrix (ECM) is a non-cellular tissue architecture containing biophysical and biochemical cues and functions as a reservoir of various biomolecules. Extracellular vesicles (EVs) are important mediators for intercellular communications containing biomolecules such as microRNAs, lipids, and proteins, and they have been considered to exist in a liquid phase. Recently, a new subpopulation of extracellular vesicles, matrix-bound nanovesicles (MBVs), was discovered and found to be bioactive. However, there is no broad proteomics-based analysis of MBVs although properties of microRNAs and lipids have been revealed. In this study, we isolated MBVs from fibroblast-derived ECM and performed LC-MS/MS-based global proteomic characterization to demonstrate how MBVs and soluble EVs differ in protein contents. We discovered that there were distinct proteomic properties of MBVs compared to soluble EVs in specific proteins, EV transport, and EV-ECM interactions. Therefore, this study is expected to provide a proteomics-basis for future studies on MBVs acting as unique microenvironment stimuli in the ECM.
세포외기질은 생물물리학적 및 생화학적 신호를 포함하는 비세포 조직 구조이며 다양한 생체 분자의 저장소 역할을 한다. 세포외소포는 마이크로RNA, 지질, 단백질과 같은 생체 분자를 포함하는 세포간 소통의 중요한 매개체로서 액상에 존재하는 것으로 여겨져 왔다. 최근에 세포외소포의 새로운 하위 집단인 기질결합 나노소포가 발견되었으며, 이는 세포외기질에서 분리되어 생물학적 활성을 나타내는 것으로 밝혀졌다. 그러나 기질결합 나노소포의 마이크로RNA와 지질의 특성은 밝혀졌지만, 광범위한 단백질체학기반 분석은 없는 상황이다. 본 연구에서, 우리는 섬유아세포 유래 세포외기질에서 기질결합 나노소포를 분리했고, 액체 크로마토그래피-텐덤질량분석법 기반의 전반적인 단백질 특성화를 수행하여 기질결합 나노소포와 액상에 떠다니는 세포외소포가 단백질 함량에서 어떻게 다른지를 보여줬다. 우리는 기질결합 나노소포가 액상에 떠다니는 세포외소포와 비교하여 특이적 단백질, 세포외소포 수송 및 세포외소포-세포외기질 상호작용에서 뚜렷한 단백질 특성이 있음을 밝혀냈다. 따라서, 이 연구는 세포외기질에서 독특한 미세 환경 자극으로 작용하는 기질결합 나노소포에 대한 향후 연구에 단백질적체학적인 기반을 제공할 것으로 기대된다.
