Microtubules (MTs) are hollow 25 nm diameter protein nanotubes comprised of dimeric alpha- and betatubulin subunits. One of the major components of the eukaryotic cytoskeleton, MTs are essentially involved in cell morphology, intracellular trafficking, and cell division. Paclitaxel (PTX) is a well known MT-targeting anticancer drug that stabilizes MT, suppressing cell division. Another MT stabilizer that is known to prevent cell mitosis is laulimalide (LMD). Although PTX and LMD both suppress MT dynamics, they attach to different binding sites of the tubulin dimer. Their influence on the structural and mechanical properties of MTs, however, is not clearly understood. We show our recent findings on the effects of PTX and LMD regulating the lattice structure using cryogenic electron microscopy (cryo-EM). To investigate the flexural rigidity of MTs, studies were conducted using transmission electron microscopy (TEM), confocal microscopy, and epi-fluorescence imaging.
마이크로튜블은 체세포 분열과 신경 세포 물질 수송에 중요한 역할을 수행하는 튜블린 단백질 나노튜브로, 여러 항암 물질들은 마이크로튜블을 표적으로 삼아 결합하여 세포분열을 억제하는 치료제로 쓰이고 있다. 그 중 대표적인 항암 물질로 쓰이고 있는 파클리탁셀은 마이크로튜블을 안정화해서 분해되는 것을 막아 마이크로튜블의 역학을 억제하는 성질이 있다. 이렇게 마이크로튜블을 안정화해서 항암작용을 할 수 있는 또다른 약물로 라우리말라이드가 있는데, 파클리탁셀과 다른 마이크로튜블 결합부위를 가지고 있다. 약물이 붙는 결합부위에 따라 마이크로튜블의 격자구조가 달라지는데, 이것이 어떻게 달라지고 어떤 영향을 주는지 나노범위에서 명확하게 밝혀져 있지 않다. 본 연구는 파클리탁셀과 라우리말라이드 약물의 영향에 따른 마이크로튜블의 나노구조를 분석하기 위해 초저온전자현미경을 이용하여 마이크로튜블의 구조를 분석하고, 휨강성을 알아보기 위해 투과전자현미경, 공초점주사 현미경, 형광 현미경을 이용하였다.
