N-Myc downstream regulated gene 3 (NDRG3) is recently emerged in physiological or pathophysiological role through mediating growth signals. Unlike NDRG1, NDRG2 or NDRG4, understanding of physiological function for NDRG3, except for elucidation as a scaffolding protein regulating growth signaling in cancer, is not enough because of limitation of germ line animal model by embryonic lethality unlike other NDRG family via knockout model. Here, I vindicates that the liver-specific deletion of NDRG3 is involved in impairment of homeostasis including flux of glucose-glycogen and hepatomegaly, and hypoglycemia. Disruption of NDRG3 in mouse liver caused extravagant glycogen synthesis with attenuating mRNA expressions of PYGL and G6PT. Moreover, metabolome analysis elucidated that NDRG3-abolished mice were ruined in balance of adenosylmethionine (SAM) and SAH induced by attenuation of glycine N-methyltransferase (GNMT) governing conversion between two metabolites as primary source of a methyl donor. Suppression of GNMT induced alteration of methionine cycle, redirecting its flux towards branch pathways to upregulate several metabolites known to have hepatoprotective functions. In addition, abrogation of NDRG3 inhibited transcriptional level of GNMT, an essential enzyme of methionine cycle, accompanying suppressed transcription of G6PT and PYGL, a rate-limiting enzyme for glycogen degradation. To summarize, these data suggest that NDRG3 is required for homeostasis of liver cell metabolism upstream of the glucose–glycogen flux and methionine cycle and provide therapeutic possibility in relation to disorders of glycogen metabolism.
NDRG3는 암의 진행에 관여한다고 보고되어 있는 scaffording 단백질로 알려져 있지만 세포 대사에서 NDRG3의 역할에 대한 연구는 미흡한 상태이며 더불어 NDRG3결손 동물의 배아 단계에서의 사망으로 인해 기능 연구에 어려움을 겪고 있다. 이런 단점을 보완하고 세포 대사에서 NDRG3의 역할을 규명하고자 간 특이적 NDRG3 결손 마우스 모델을 처음으로 제작하였다. NDRG3 결손 마우스는 글리코겐 축적 질환(GSD)에서 나타나는 간 손상을 동반하는 글리코겐의 비정상적 축적, 간의 비대화, 저혈당 증상을 수반하였다. 글리코겐의 비정상적 축적을 보이는 NDRG3의 결손 마우스는 글리코겐 합성의 핵심 유전자로 알려진 PYGL과 G6P의 수송을 담당하는 유전자인 G6PT 유전자들의 발현 감소를 동반되었다. 대사체 분석을 통해 간에서 NDRG3 결손으로 인해 메티오닌 회로의 왜곡이 발생되고 연속적으로 메티오닌 회로에 커플링된 주변 신호전달 체계들도 손상됨을 확인하였다. 특히 SAM, MTA, 시스타티오닌 그리고 글루타치온 같은 방어적 기작을 위한 메티오닌 회로 관련 대사물질들의 증가가 관찰되었다. 분자기전 측면에서 메티오닌 회로에서 SAM을 SAH로 전환하는 GNMT의 양적 저하가 SAM의 양적 증가를 유도하는 주된 원인임을 확인했다. 또한 NDRG3의 결손에 의해 야기된 GNMT의 감소는 글리코겐 분해에 관련된 PYGL과 G6PT의 저해를 동반함을 확인하였다. 요약하면 본 연구를 통해 NDRG3가 간세포에서 글리코겐과 포도당의 전환 및 메티오닌 회로의 상위 조절에 요구되며 또한 이로 인한 글리코겐 대사 관련 질환의 치료적 가능성을 제시하였다.
