Over the past decade, data-driven reconstruction of gene regulatory network has been an important subject of study as a clue to the systems level understanding of the biological phenomena, but there was limitation on the amount of data available. However, developments in single cell technology and pseudotime analysis allowed single cell data regarded as time-series data. Especially, algorithm to infer the network with dynamics are being developed, beyond the reconstruction of network topology alone. In this thesis, a Boolean network reconstruction method based on pseudotime analysis of high-dimension single-cell RNA-seq data is proposed, and applied to the single-cell data of early colon cancer progression data. The reconstructed network is then analyzed if it can provide a systems level understanding of the tumorigenesis.
지난 십수 년간 데이터 기반의 유전자 조절 네트워크 재구축은 생명현상을 시스템 관점으로 이해할 수 있는 단서로서 주요한 연구 주제로 다루어져 왔으나, 얻을 수 있는 데이터 양에 의한 한계가 존재하였다. 그러나 최근 단일세포 기술의 발달과 유사시간 분석의 개발로 단일세포 데이터를 시계열 데이터와 같이 다룰 수 있게 되어 이러한 한계를 극복하고 있으며, 특히 기존의 네트워크 구조만의 재구축을 넘어 유전자 간 조절관계가 포함된 동역학 네트워크를 추론할 수 있는 알고리즘의 개발이 이루어지고 있다. 본 학위논문에서는 고차원 단일세포 전사체 데이터의 유사시간 분석을 통한 불리언 네트워크 모델 구축 방법을 다루고자 하며, 해당 방법을 초기 대장암 발생 과정을 모사하는 단일세포 데이터에 적용하여 모델을 구축하여, 구축된 모델이 암화과정에 대한 시스템 차원의 이해를 제공할 수 있는지 분석해보고자 한다.
