The recent advent of connectomics has provided us with a huge amount of brain network data as well as a new way of understanding complex brain functions in view of network science. Recently, applying network control theory to brain networks has become an emerging issue, and the significance of network controllability in the brain has also been validated based on clinical data. Since the brain itself is already an autonomously controlled system which governs the core functions of life at all times, it can be inferred that the control architecture of brain networks should have a certain degree of robustness in maintaining normal brain functions. In this regard, analysis on robustness of the network controllability in the brain might provide a novel insight into the inherent control architecture of the brain and lead us to a deeper understanding of the organizational principle of the brain. However, there was no such an attempt. To tackle this problem, in this thesis, I have extracted structural brain networks of 100 healthy adults from the Human Connectome Project database, and also prepared the structural brain networks of other species for comparison. I have simulated various perturbations on brain networks and analysed the resulting change of network controllability metrics. Furthermore, I compared these results with those obtained from corresponding null networks, canonical model networks, and other types of real-world complex networks. As a result, I found that brain networks have stronger robustness in their network controllability with respect to random and targeted attack. I also found that these results originate from the inherent control architecture of brain networks, which is different from other types complex networks. The findings in this thesis may provide a more advanced understanding of the operating principle of brain functions and contribute to designing a more effective therapeutic intervention for cognitive enhancement and treatment of complex brain disorder.
최근 커넥토믹스(connectomics)의 부상으로 인해 다양한 종류의 브레인 네트워크 데이터들을 다량으로 확보할 수 있게 되면서, 네트워크 과학의 관점에서 복잡한 뇌의 기능을 해석하고자 하는 새로운 연구들이 활발히 진행되고 있다. 그 중에서도 네트워크 제어 이론의 응용은 근래에 많은 주목을 받고 있는데, 이러한 방법론의 유효성이 다수의 임상 실험들을 통해서 확인되고 있다. 시스템 제어 이론의 관점에서 생명체의 뇌신경 시스템은 생체 내의 핵심적인 기능들을 수행하고 조정하는 기관이므로, 정상적인 동작 수행을 위해 충분한 제어적 강건성을 갖추고 있을 것이라 유추해 볼 수 있다. 따라서 브레인 네트워크의 제어적 강건성을 분석하는 것은, 브레인 네트워크에 내재된 제어 구조를 파악하고 뇌의 설계 원리에 대한 보다 깊은 이해를 이룰 수 있다는 점에서 중요하다고 할 수 있다. 따라서 본 학위논문에서는 인간을 포함한 여러 종의 브레인 네트워크들을 구축하고, 이들을 대상으로 제어적 강건성 분석을 실시하였다. 그리고 이러한 결과들을 널 네트워크, 모델 네트워크 및 현실의 다양한 네트워크들과 비교하여 브레인 네트워크의 제어적 강건성이 갖는 특징을 분석해 보았다. 이를 통해 브레인 네트워크는 제어적 관점에서 무작위적인 공격 패러다임과 연결성 기반의 공격 패러다임에 대해 상대적으로 강건한 특성을 보였으며, 이것이 브레인 네트워크에 내재된 제어 구조에서 유래함을 확인할 수 있었다. 본 학위논문에서 수행한 분석들은 뇌의 동작과 설계 원리에 대한 정밀한 이론을 구축하는데 기여할 수 있을 것이며, 복잡한 뇌 질환의 치료전략을 제시하고 인지기능의 향상을 위한 기술 개발에 기초를 제공할 수 있을 것으로 기대된다.
