Biochemical processes in cells are proceeded in various organelles. Recently, diverse membrane-less organelles (MLOs) are reported in cell, which are known to have important roles in many biochemical processes of cell. MLOs are constructed by liquid-liquid phase separation (LLPS), which drives compartmentalization and generation of liquid droplets without physical barriers. Various proteins are incorporated in the liquid droplets, and many of them are known as intrinsically disordered proteins (IDPs), which have partial or overall disordered regions inside. Importantly, multivalent interactions of IDPs are known as a key-step of LLPS, and the proteins amino acid proportion is known as a significant factor to determine droplets physical properties. However, how potential interactions between different IDPs affect the behavior of protein droplets is largely unknown. In this study, we develop rapid IDP clustering system to generate protein liquid droplets with varied residue compositions and examine diverse interacting IDPs inside and outside of droplets. Additionally, we investigate molecular affectances for tendency and power of protein recruitment into the protein liquid droplets.
세포 내의 생물·화학적 반응들은 다양한 세포 소기관 안에서 진행된다. 근래에는 세포 내에 다양한 막 없는 소기관 (Membrane-less organelles)들이 보고되었고, 이들이 생명 활동에서 중요한 역할을 한다는 사실이 밝혀지고 있다. 막 없는 소기관들은 물리적 장벽 없이 액체 상분리 현상 (liquid-liquid phase separation)에 의해 구획화 (Compartmentalization)되어 방울형 구조체 (Liquid droplet)를 형성한다. 구조체 안의 다양한 단백질 중 많은 수가 고정된 구조를 갖지 않는 비정형 단백질 (Intrinsically disordered proteins)임이 알려져 있으며, 이들은 다중 상호작용 (Multivalent interaction)을 통해 액체 상분리 현상을 유도할 수 있다. 하지만 서로 다른 비정형 단백질 사이의 상호작용이 단백질 구조체의 성질에 미치는 영향에 대해서는 많이 알려져 있지 않다. 본 연구에서는 비정형 단백질을 군집화 (Clustering)하여 상분리 현상을 일으키는 시스템을 만들고, 다양한 단백질로 만든 방울형 구조체가 다른 단백질 혹은 구조체와 어떻게 상호작용하는지 관찰하였다. 또한 단백질이 구조체 내부로 모집 (Recruit)되는 과정에서, 어떤 조건이 그 경향성과 세기에 영향을 미치는지 알아보고자 하였다.
