In endothelial cells (ECs), differential vascular endothelial growth factor receptor 2 (VEGFR2) signaling in a nascent sprout yields two distinct cellular phenotypes, tip ECs and stalk cells, in a stochastic pattern. However, the precise mechanisms underlying how intracellular VEGFR2 signaling is higher in one EC compared with its neighbour and acquires the tip EC phenotype under a similar external cue are elusive. Here, we show that Merlin, encoded by the neurofibromatosis type-2 (NF2) gene, suppresses VEGFR2 internalization depending on vascular endothelial cadherin (VE-cadherin) density and inhibits tip EC induction. Accordingly, endothelial Nf2 depletion significantly promotes tip EC induction with excessive filopodia by enhancing VEGFR2 internalization not only in the growing vessels but also in the fully matured vessels. Mechanistically, Merlin physically binds to the VEGFR2–VE-cadherin complex at cell–cell junctions and reduces the activity of VEGFR2 internalization–induced downstream signaling pathways during tip EC induction. As a consequence, non-functional excessive sprouting largely occurs during tumor angiogenesis in EC-specific Nf2-deleted mice, leading to delayed tumor growth. Together, our findings show that Nf2/Merlin is a crucial molecular gatekeeper for tip EC induction, capillary integrity, and proper tumor angiogenesis by suppressing VEGFR2 internalization.
혈관 신생은 혈관내피세포의 활동이 짜임새있게 조절되는 생명현상이다. 혈관 신생이 일어날 때 혈관내피세포가 vascular endothelial growth factor receptor 2 (VEGFR2) 신호전달체계를 통해 tip cell과 stalk cell로 분화가 된 후 이동, 분열, 세포의 성숙 등이 일어난다. Tip 세포는 filopodia를 내면서 이동해야하는 방향을 결정하는 반면 stalk 세포는 tip 세포 뒤에 따라오는 세포로, proliferating을 통해 lumen을 형성한다. 이러한 Tip 과 stalk 세포 분화에 대해서는 현재까지 연구가 활발히 이루어졌다. 하지만 비슷한 외부 신호 아래에서 무작위 패턴으로 특정 혈관 내피세포가 다른 옆 세포들보다 VEGFR2 signaling이 높아지면서 tip 세포로 분화가 되는데 어떠한 메커니즘으로 특정 혈관내피세포가 tip 세포로 선택 되는지에 대해서는 밝혀진 바가 없다. 본 연구에서는 tumor suppressor로 알려져 있는 Merlin (유전자 이름은 Nf2)이라는 단백질이 VEGFR2 신호전달체계를 통해 혈관 생성에서 일어나는 tip cell induction을 조절하는 메커니즘을 규명하였다. Merlin이 vascular endothelial cadherin (VE-cadherin) 밀도에 따라 VE-cadherin과 VEGFR2와의 결합으로 VEGFR2 internalization을 조절함으로써 tip cell induction이 조정된다는 것을 밝혔다. 혈관내피세포 특이적으로 Nf2를 없앴을 때 자라나는 혈관뿐만 아니라 adult 마우스의 mature한 혈관에서도 VEGFR2 internalization이 증가함으로써 filopodia와 tip 세포 수가 wild-type(WT)에 비해 증가하는 것을 확인하였다. 본 연구결과는 혈관신생과정에서 Nf2/Merlin의 역할을 밝힌 것 뿐만 아니라, tip cell 분화에 있어 정확한 메커니즘을 규명하였다.
