Microbiota is essential in development and functional maturation of our immune system, even outside the gut. However, whether and how gut microbiota may affect skin myeloid cells is unclear. Present study presents a microbiota-dependent macrophage subset that aid in protection against skin infection. I found evident downregulation of type I interferon signaling in skin of germ free mice compared to specific pathogen free mice. The decrease in type I interferon signaling was closely related to microbiota presence and a macrophage-specific marker Siglec-1 (CD169). CD169 expression was inducible using recombinant type I interferons in bone marrow-derived macrophages. Absence of CD169$^+$ macrophages resulted in increased bacterial burden and impairment of immune response against S. aureus skin infection. CD169$^+$ macrophages mediated not only recruitment of neutrophil and γδ T cells, but also activation of γδ T cells via IL-23. Absence of CD169$^+$ macrophages lead to diminished number of IL-17 producing T cells, which are essential for host defense against S. aureus. My findings verify a role of microbiota in establishment of a specific myeloid cell subset expressing CD169 in the skin and provide specific mechanism of how the subset protects against bacterial skin infection.
공생미생물은 피부 면역에 다양한 방법으로 관여하는 것으로 알려져 있다. 그러나 공생미생물의 피부내재골수성세포에 대한 영향은 그 연구가 미비한 실정이다. 본 연구에서는 공생미생물이 조직내재대식세포의 표지자중 하나인 Siglec-1(CD169)에 영향을 주는 기전을 확인하였다. 적으나 지속적인 량의 1형 인터페론 분비는 피부 조직의 항바이러스 면역 작용에서 더 나아가 CD169$^+$ 대식세포를 피부에 존재하게 한다. CD169$^+$ 대식세포가 없는 쥐에 피부 감염을 시켰을 때 더 큰 피부 병변과 높은 균 역가를 보였다. CD169$^+$ 대식세포는 피부의 진피 아랫부분과 피하지방의 혈관 주위에 존재한다. 황색포도알구균의 진피 피부 감염시 CD169$^+$ 세포는 호중구를 모집하여 초기 면역 반응에 중요한 역할을 하며, 이후에는 γδ T 세포의 모집과 활성화에 중요한 역할을 하여, 황색포도알구균의 퇴치에 필수적인 역할을 한다. CD169는 자극을 통하여 조절할 수 있는 세포접착분자이므로, 이에 대한 연구를 통해 감염 치료 및 방지에 새로운 방안을 제시할 수 있으리라 생각된다.
