Ischemic diseases such as myocardial infarction and cerebral infarction are caused by the restriction in blood supply to tissues and can be treated by early reperfusion. However, reperfusion injury is observed even though the reperfusion is performed to restore blood flow if the ischemia is maintained for a certain period. To analyze changes in blood flow and mechanical properties in tissues, optical coherence tomography (OCT) was used. OCT is an imaging technique using light interference which can provide the capability of non-invasive imaging over a wide field at high speed, thus OCT is widely used for biomedical and clinical applications. In this thesis, a multi-functional OCT system allowing the visualization of changes in blood flow and tissue mechanical properties was developed to study murine ischemia and reperfusion models. A new imaging protocol based on OCT imaging was proposed to measure the change of flow direction occurred during ischemia and reperfusion process.
심근경색이나 뇌경색과 같은 허혈성 질환은 조직으로의 혈류 공급이 차단되어 발생하는 질병으로 빠른 재관류를 통해 치료가 가능하다. 하지만 일정 시간 이상 허혈 상태가 유지될 경우, 재관류를 시켜 혈류를 회복시켜주더라도 조직의 손상이 진행되는 재관류 손상이 관찰된다. 특정 조직에서의 혈류와 기계적 성질의 변화를 분석하기 위하여 광간섭단층촬영 기법을 사용하였다. 광간섭단층촬영은 빛의 간섭 현상을 이용한 영상 기술로 비침습적으로 생체 조직의 넓은 영역을 고속 영상화할 수 있어 생의학 및 의학 분야에서 연구 및 질병 진단에 널리 활용되고 있다. 본 학위논문에서는 조직의 혈류와 기계적 특성의 변화를 영상화할 수 있는 다기능 광간섭단층촬영 시스템을 개발하여 쥐 허혈 및 재관류 모델에 대하여 적용하였다. 허혈 및 재관류 과정에서 관찰되는 혈류 방향의 변화를 측정하기 위한 광간섭단층촬영 기법 기반의 새로운 이미징 방법을 제시하였다.
