The object of this project is to find W.B.C. in the blood sample, focus blood sample image using electrical signal converted by TV camera, digitize this signal by 64×64 pixels, store the digitized data in RAM, preprocess into background, cytoplasm. and nucleus, extract features in BIP (binary image plane) and classify the type of W.B.C.
Of the above objects, this system describes the process of autofocusing and searching.
This process is as follows.
First, blood sample magnified through microscope by 1000 is changed to electrical signal by Color TV camera.
This electrical signal is composed of blue color, red color and gray signals. In order to search, blue signal is amplified by 100 and to focus, gray signal is digitized by high speed A/D converter (TDC 1007J).
For this functions, it is necessary to move microscope in the X, Y and Z directions.
Three stepper motors (SIGMA INSTRUMENTS) are used for moving microscope in all directions. X-Y table is used to lay blood sample slide and it, is moved automatically by X, Y stepper motors. Z coupler is used to couple Z motor and microscope.
The microcomputer that uses Z-80 CPU controls searching and focusing. In this system, in searching, the pulse which indicates W.B.C. interrupts Z-80 CPU and the data around W.B.C. transfers digitizer and then it is searched again. The method to focus in this system is that the gray level above threshold is added and the summed data transfers Z-80 CPU, and it moves Z-stepper motor in the direction of maximizing the summed value.
The control program which programmed according to above algorithm is stored in 2716 EPROM. In this way, above system is designed and made up.
본 System은 백혈구 자동류기 proiect중 W.B.C Search와 현미경 Focus를 다루었다.
우선 blood sample을 현미경 위에 놓고 이를 TV Camera를 통하여 전기적인 signal을 얻는다.
이 signal을 사용하여 W.B.C를 search하고, window position을 digitizer로 보낸다.
digitizer와 High Speed A/D converter를 통과한 8 bits data를 사용하여 Focus하였다.
Search를 위하여 X , Y stepper motor를 사용하였고, Focus를 위하여서는 Z stepper motor를 사용하였다.
이 Search와 Focus를 위하여 움직이는 X, Y, Z Stepper motor는 Z-80 SBC에 의하여 control되었다.
