One thousand thermophiles isolated from soils were screened for hydantoinase and its thermostability. One thermophilic bacterium that showed the highest thermostability and activity of hydantoinase was identified to be Bacillus stearothermophilus SD-1 according to morphological and physiological characteristics.
Culture conditions for the production of thermostable D-hydantoinase from thermophilic Bacillus stearothermophilus SD-1 were investigated. Yeast extract was most effective as a nitrogen source, and the addition of carbon sources gave no significant increase in enzyme formation. The production of D-hydantoinase was repressed by the presence of glucose during the cultivation, but, when glucose was depleted, the enzyme activity remarkably increased. Manganese ions were found to be essential, but no inducer was required for the production of D-hydantoinase from B. stearothermophilus SD-1. The microorganism showed a linear growth in the medium containing yeast extract, and addition of meat extract enabled an exponential growth. The D-hydantoinase of B. stearothermophilus SD-1 was synthesized at the stationary phase in the medium supplemented with meat extract, and enzyme production level was improved about three times compared to that in the absence of meat extract.
The thermostable D-hydantoinase of thermophilic B. stearothermophilus SD-1 was purified to homogeneity by using an immunoaffinity chromatography. The affinity chromatography using polyclonal antibody immobilized on Sepharose 4B was relatively simple to operate and gave a purification yield of 60%. Molecular mass of the enzyme was determined to be about 133.9 kDa on gel filtration chromatography and the molecular mass of the subunit was 54 kDa on SDS-PAGE. Mass spectrometer was also applied to the determination of molecular mass, and molecular masses of the subunit of enzyme and native enzyme were obtained as 50.1 and 102.1 kDa, respectively. From the molecular mass determined by two methods, the D-hydantoinase is presumed to be organized as a dimer. Stokes radius and friction ratio of the enzyme are calculated as 4.34 nm and 1.40, respectively. Isoelectric pH of the enzyme was observed to be 4.47. The enzyme required manganese ions for the activity, and manganese content was found to be one manganese ion per molecule of enzyme. The optimum pH and temperature were about 8.0 and 65℃, respectively, and the half-life of the enzyme was estimated to be 30 min at 80℃, indicating the most thermostable enzyme so far. Kinetic constants of the enzyme for different substrates were also determined.
The enzyme was immobilized on various support matrix by adsorption, and DEAE-cellulose resin was found to be most effective in terms of the activity recovery and the amount of protein bound. The activity of enzyme immobilized on DEAE-cellulose was retained more than 80%, and the optimal reaction conditions for the immobilized enzyme were determined to be 55℃ and pH 9.0, respectively. Immobilized enzyme was applied to the production of N-carbamoyl-D-p-hydroxyphenylglycine from D,L-5-(4-hydroxyphenyl) hydantoin in repeated batch, and the production rate was maintained constantly over 9 successive operations.
A gene encoding D-hydantoinase of B. stearothermophilus SD-1 was cloned in E. coli. B. stearothermophilus SD-1, chromosomal DNA fragment (10.5 kb) which was expected to contain the D-hydantoinase gene was previously inserted in pBR322 and constructed a new plasmid pHBR5. By subcloning and restrictive size-deletion of the 10.5 kb insert, a pUC18 plasmid containing a 3.1 kb insert was constructed and designated pHU183. The D-hydantoinase was overproduced in E. coli strain containing the plasmid of pHU183. With SDS/PAGE analysis, the content of D-hydantoinase was found to be about 20-30% of total protein in E. coli. The D-hydantoinase could be produced efficiently without IPTG-induction in LB medium.
고온성 미생물이 서식 할 수 있는 온천수, 고온성 폐수, 두엄, 비닐하우스, 얕은 개울바닥 및 부식토 등에서 55℃에서 생육가능한 천 여개의 고온균을 분리하여, hydantoinase 활성과 열안정성을 조사하였다. 그 결과, 탁월하게 높은 열안정성을 보인 한 균주를 선별하였으며, 형태 및 생리학적 동정에 의하여, 이 균을 Bacillus stearothermophilus SD-1으로 분류하였다.
이 고온균으로부터 열안정성 hydantoinase를 생산하기위한 최적배지조건 및 발효적 특성에 관한 연구를 수행하였다. 이 균은 효모추출물을 영양원으로 공급하였을 경우에 가장많은 hydantoinase를 생산하였고, 탄소원들은 효소생산에 거의 영향을 미치지 않았다. 특히, 포도당을 넣은 배지에서 이 균을 배양하면, 효소생산이 억제되다가 배양액 중의 포도당이 고갈된 후에 갑자기 증가하는 특성을 보였다. 일반적으로 알려진 바와는 달리, 기질유도체의 첨가에 따른 효소생산의 증가는 거의 없었고, 망간 금속이온은 균 증식에는 영향을 미치지 않았으나 효소생산에 필수적이었다. 이 고온균은 효모추출물을 영양원으로 배양 할 때, 초기부터 선형적으로 증식하는 비정상적인 성장곡선을 보였다. 비타민이나 금속이온 들은 선형적 증식에 영향을 미치지 않았고, 고기추출물을 배지에 첨가하였을 때, 비로소, 이 고온균은 정상적으로 대수증식하였다. 이와같이 고기추출물을 배지에 첨가하여 고온균이 정상적으로 증식하였을경우, 효소생산량이 약 세배 증가하였다.
이 고온균이 생산하는 hydantoinase의 성질을 밝히기 위하여 순수정제하였다. 먼저, 전통적인 chromatography 방법으로 부분정제 한 뒤, preparative gelelectrophoresis 방법으로 이 효소를 순수분리하였다. 그리고, 이와같이 순수분리된 단백질을 토끼에 주입하여 이 효소에 특이적으로 결합하는 항체를 생산 한 후, 이 항체를 담체에 고정화하여, 단 한 번의 친화성 chromatography로 hydantoinase를 정제하는 방법을 개발하였다.
이 효소의 분자량은 gel filtration chromatography 방법에 의하여 133,000으로 측정되었고, subunit의 분자량은 SDS/PAGE 방법에 의해 54,000으로 측정되었다. 분자 질량 분석기로 이 효소를 분석하였을 경우에는, 이 효소와 subunit의 분자량이 각각 102,100과 51,050으로 결정되었다. 여러가지 방법으로 측정한 분자량으로부터, 이 효소가 같은 크기의 두 분자가 결합하여 존재하는 단백질인 것을 유추 할 수 있었다. 이 효소의 Stokes radius 와 friction ratio는 각각 4.34 nm 와 1.40으로 계산 되었다. 이 효소는 효소의 작용에 망간금속이온을 반드시 필요로 하였는 데, 효소분자 하나에 한 분자의 망간이온을 포함하고 있었다. 이 효소는 hydantoin 유도체의 D-isomer에만 특이적으로 작용하였다.
효소작용을 위한 최적 pH 와 온도는 각각 8.0 과 65℃ 였고, 반감기는 80℃에서 30 min 이었다. 이 열안정성은 현재까지 보고된 hydantoinase 중 가장 높은 것이다. 이 효소의 Michaelis 상수 ($K_m$) 는 dihydrouracil과 hydantoin에 대하여 각각 1.1 mM과 340 mM로 결정되어, 이 효소가 세포 내에서는 dihydropyrimidine의 대사에 관여하는 효소인 것으로 판단된다.
이 효소를 써서 D,L-p-hydroxyphenylhydantoin로부터 N-carbamoyl- D-hydroxyphenylglycine을 생산하기 위한 연구를 수행하였다. 여러 고정화 담체를 비교한 결과 DEAE-cellulose가 효소활성 회수율 (80%)이나 고정화된 단백질의 량 (15 mg/ml)으로 볼 때, 가장 우수하였다. 이 반응을 효율적으로 수행하기 위한 온도 및 pH 조건은 각각 55℃ 와 pH 9.0으로 결정되었다. 이 고정화효소를 이용하여 앞서의 효소적 전환반응을 효소활성의 손실없이 아홉번 반복 할 수 있었다.
B. stearothermophilus SD-1의 D-hydantoinase를 encode 하는 유전자를 대장균에 cloning 하였다. 이 유전자를 플라스미드 pBR322에 삽입하고, 이 플라스미드를 pHBR5로 이름지었다. pHBR5에 들어간 고온균 유래의 유전자를 제한효소로 잘라서 크기를 3.1 kb로 줄이고 플라스미드 pUC18에 삽입하였다. 이와같이 얻어진 새로운 플라스미드를 pHUC183으로 이름지었다. pHU183을 포함한 대장균을 LB 배지에서 배양하였을 때, hydantoinase 생산량은 원래 균주에서 보다 약 10배 이상 증가하여 대장균이 생산하는 총단백질의 약 20%가 이 효소인 것으로 밝혀졌다. 특히 배양액에 IPTG를 첨가하지 않는 경우에도, 비슷한 수준으로 효소가 생산되었다.
