Microfluidic technologies have a number of advantages for sample preparation and analysis in point-of-care testing (POCT), but the needs of external pumping systems limit the wide use of them. To eliminate the needs of complicated external pumping systems, various power-free microfluidic devices have been developed. However, it is still challenging for on-demand control of various flow mechanisms without the use of any external equipment. In this thesis, I present a finger-actuated microfluidic device based on indirect pressurization to control a set amount of fluid regardless of differences in end users. Using a diaphragm, active and passive check valve, not only dispensing, but also reciprocation, flow direction control, and mixing of multiple reagents was demonstrated and each flow behavior was characterized. By applying the proposed flow mechanisms, I developed the devices for practical microfluidic sample preparation and analysis in POCT. First, the devices for blood typing and cross-matching test was developed as pre-transfusion tests to prevent hemolytic transfusion reaction. As rapid pre-transfusion tests for urgent transfusion patients, the results can be simply obtained by pushing the button. A smart blood typing system was introduced that displays the letters of blood type as a first-line test, and a fully-integrated cross-matching test was demonstrated as a final verification. Second, conventional solid-phase nucleic acid purification procedure was simplified using the finger-actuated microfluidic devices. The reagents are sequentially delivered to the silica bead column via a predesigned fluidic path according to the actuated button to obtain purified nucleic acid. A reciprocating flow was further assisted to enhance the recovery rate of nucleic acid. As a target analyte, standard Hepatitis B virus (HBV) DNA was successfully purified with up to 50% of the recovery rate. Third, the proposed device was conducted to substitute accurate manual pipetting or robotic arms required for conventional biological analysis. As a potential application, a finger-actuated microfluidic concentration gradient generator was developed for studying enzyme kinetics, and the device was utilized for spheroids culture and analysis in hanging drop array. In addition to applications presented in this thesis, the proposed device can be widely used as a robust tool to facilitate sample preparation and analysis in POCT, and conventional biological analysis.
미세 유체 기술은 POCT (point-of-care test)에서 시료 준비 및 분석 과정을 용이하게 하기 위한 다양한 장점들을 가지지만, 미세 유체 제어를 위한 외부 펌핑 시스템의 필요성으로 인해 널리 사용되지 못하였다. 복잡한 외부 펌핑 시스템의 필요성을 없애기 위해 다양한 무전원 미세 유체 장치가 개발되었지만, 외부 장비를 사용하지 않고 다양한 유체의 흐름을 주문형으로 제어하는 것은 여전히 어려운 과제로 남아있다. 이 학위 논문에서는, 최종 사용자에 관계없이 일정량의 유체 흐름을 제어하기 위해 간접 가압 방식에 기반한 손가락 작동 식 미세 유체 장치를 제안한다. 간접적으로 작동하는 다이어프램과 능동 및 수동 역류 방지 밸브를 사용하여 유체의 분배 뿐만 아니라 왕복 운동, 방향 제어 및 여러 시약의 혼합을 시연하고 각 흐름 특성을 분석하였다. 제안 된 작동 원리를 적용하여 POCT를 위한, 미세 유체 기술에 기반한 실용적인 시료 준비 및 분석용 장치들을 개발하였다. 먼저, 용혈성 수혈 부작용을 방지하기 위한 수혈 전 검사들로 혈액형 검사 및 교차 적합 시험을 위한 장치들을 개발하였다. 응급 수혈이 필요한 환자를 위한 신속한 수혈 전 검사로서, 버튼을 누르는 간단한 작동 만을 통해서 수혈 적합성을 확인할 수 있다. 가장 처음 이루어 져야 하는 수혈 전 검사로, 혈액형에 해당하는 문자를 미세유체채널을 통해 표시하는 스마트 혈액형 검사 방법과 수혈 적합성의 최종 판정을 위한 완전 통합 형 교차 적합 시험 법을 개발하였다. 두번째로, 기존의 복잡한 고상 핵산 정제 방법을 손가락 작동 방식의 미세 유체 장치를 사용하여 단순화 하였다. 시약들은 작동 버튼에 따라 미리 설계된 유체 경로를 통해 실리카 비드 컬럼으로 순차적으로 전달되어 핵산을 정제하였다. 핵산의 회수율을 향상시키기 위해 왕복 유동이 추가적으로 사용되었다. 표적 분석 물질로써, 표준 B 형 간염 바이러스 DNA를 약 50 %의 회수율로 성공적으로 정제하였다. 세번째로, 제안 된 장치는 기존의 생물학적 분석에 필요한 정확한 수동 피펫팅 또는 로봇 팔을 대체하기 위해 사용되었다. 잠재적인 응용분야로, 효소 동역학을 연구하기 위한 손가락 작동 방식의 미세 유체 농도 구배 생성기가 개발되었다. 또한, 손가락 작동 방식의 미세 유체 장치는 매달린 방울 배열에서 스페로이드 배양 및 분석을 용이하게 하기 위해 사용되었다. 본 학위논문에서 제시한 적용 분야 외에도 제안 된 장치는 POCT의 시료 준비 및 분석과 기존의 생물학적 분석을 용이하게 하기 위한 강력한 도구로서 널리 사용될 수 있을 것이다.
