5-Aza-2'-deoxycytidine, also known as decitabine, is a DNA hypomethylating agent (HMA) used to treat acute myeloid leukemia (AML) and pre-leukemic disorder myelodysplastic syndrome (MDS). Decitabine induces cell death by activating transcription of endogenous retroviral elements (ERVs). When transcribed, ERV RNAs can adopt double-stranded secondary structure and activate melanoma differentiation-associated protein 5 (MDA5), a member of the innate immune response proteins that recognize viral double-stranded RNAs (dsRNAs). Here, we employ an image-based RNAi screening system to identify dsRNA-binding factors that affect cellular response to ERV induction by decitabine. We find that Staufen1 (Stau1) knockdown decreases the interferon signature and rescues decitabine-mediated cell death. Our subsequent analyses reveal that Stau1 directly binds to ERV RNAs and stabilizes the RNAs together with a long noncoding RNA, TINCR. Analysis of a clinical patient cohort further supports that MDS and AML patients with low Stau1 expressions exhibited poor response to HMA. Our study reveals that decitabine-mediated cell death is a consequence of complex interactions among different dsRNA binding proteins for access to common dsRNA targets.
데시타빈으로 알려진 5-아자-2'-데옥시사이티딘은 DNA 탈메틸화제(HMA)로써 급성 골수성 백혈병(AML)과 백혈병 전 골수이형성 증후군(MDS)의 치료제로 사용된다. 데시타빈은 내인성 레트로바이러스(ERV)의 전사를 활성화시킴으로써 세포사멸을 유도한다. ERV는 전사되면서 이중가닥의 2차구조를 형성하고 이중가닥 RNA와 결합하는 선천적 면역반응 단백질 중 하나인 흑색종분화관련단백질(MDA5)을 활성화시킨다. 본 학위논문에서는 데시타빈에 의해서 증가하는 세포내 ERV에 영향을 미치는 이중가닥 RNA 결합 단백질을 확인하기 위해 이미지 기반의 RNA 간섭 스크리닝 시스템을 사용하였다. Staufen1을 녹다운 시켰을때, 인터페론 시그니처를 감소되는 것을 확인하였고 데시타빈에 의한 세포사멸이 회복되는 것을 확인하였다. 또한 실험을 통해 Stau1이 ERV와 직접 결합하고 긴 비-코딩 RNA 인 TINCR과 함께 ERV를 안정화시키는 것을 확인하였다. 임상 환자 코호트 분석을 통해서 낮은 Stau1 발현을 가지는 MDS와 AML 환자는 HMA에 약한 치료반응을 보인다. 본 연구는 데시타빈에 의한 세포사멸이 다른 이중가닥 RNA 결합 단백질의 복합적인 상호작용임을 보여주며 이중나선 RNA의 표적에 대한 일반적인 접근이 될 것임을 기대한다.
