Nowadays, nucleic acid based detection methods are widely reported. Most of all, molecular beacons, which fluorophore and quencher are connected to each end of hairpin structure, are mainly used. They shows a linear fluorescence signal increment as the biological target concentration increases. Here in, a detection system showing a step-wise function of fluorescence signal along the biological target concentration was developed by decoupling the fluorophores and the quenchers. The modified hairpin probes, form of hairpins with quenchers at the one end, were functionalized to polystyrene nanoparticles, and the fluorophores were loaded onto the particles. Using the newly developed probes, biological targets were successfully detected showing a step-wise function. Then, an attempt was made to improve the detection efficiency of the probe by inserting the tail of adenosines, and the effects of the probe’s fluorophore and quencher concentration on the detection profile were observed.
최근 들어 대부분의 생물학적 타겟은 핵산을 기반으로 하여 검출한다. 주로 헤어핀 구조의 양 끝에 형광 분자와 소광제가 연결되어 있는 분자 비콘을 이용하며, 생물학적 타겟 농도가 증가함에 따라 형광 신호가 선형적으로 증가하는 형태를 보인다. 따라서 본 연구에서는 형광 분자와 소광제를 분리시켜 형광 신호가 계단식으로 증가하는 검출 시스템을 구축하였다. 한쪽 끝에만 소광제가 연결된 헤어핀 탐침을 폴리스티렌 나노 입자에 기능화하고, 나노 입자에 형광 분자를 로딩하여 본 탐침을 개발하였다. 구축한 탐침을 이용하여 생물학적 타겟을 검출하였을 때, 타겟 농도가 증가함에 따라 형광 신호가 계단식 형태로 증가하는 것을 확인하였다. 이 후, 아데노신 꼬리를 삽입하여 본 탐침의 검출 효율을 높이고자 시도하였으며, 탐침의 형광분자와 소광제 농도에 따른 검출 형태 변화를 관찰하였다.
