Microelectrode array is the pivotal platform in neural science and engineering because of its ability to read out the electrical activity of living neurons. The recent development of high-density and highly scalable electrode arrays enables sub-cellular recording and wider coverage, respectively. However, it is extremely challenging to achieve both metrics in a single array. The speed requirement in highly scalable arrays induces wideband-noise folding, and this property precludes meeting the strict noise requirements in high-density arrays.
Previous studies contended that circuit thermal noises should be reduced to improve the signal-to-noise ratio of the recording system. By investigating the cell-electrode interface, however, we showed that the sealing resistance between a cell and an electrode turns out to contribute over 90% of total wideband noise power. In line with this finding, wideband-noise canceling strategy was presented to cope with the condition and support reliable recording. The recording system supported full-frame scanning of 100,000 electrodes with a $10- \mu m$ pitch. As a result, an 85% reduction of the noise contribution by the sealing resistance was achieved. Input-referred noise was reduced from $80 \mu V_{rms}$ to $18 \mu V_{rms}$. Spikes in weak-sealed cases that could not be detected without the cancellation were disclosed, and signal-to-noise ratio was significantly increased for all sealing conditions.
미세전극 어레이는 체외와 체내에서 신경 세포의 전기적 발화 정보를 읽어내는 기구로써 신경과학 및 공학계의 핵심 플랫폼으로 작용한다. 이 중에서도 최근 등장한 초 고밀도, 초 고집적 전극 어레이는 각각 서브-셀 단위의 측정과 더욱 넓은 영역의 셀 네트워크 측정을 실현하고 있다. 그러나 고밀도와 고집적 조건을 동시에 만족시키기 위해서는 큰 어려움이 따른다. 고집적 전극 어레이 스캐닝 회로에서 필요로 하는 빠른 리드아웃 속도는 고밀도 전극 어레이 회로에서 요구하는 잡음 성능을 크게 훼손시킨다. 이 때문에 현재 $10 \mu m$ 간격 고밀도 조건에서 동시에 읽어낼 수 있는 전극의 수는 수천 개 수준에 머물러 있다.
기존 연구에서는 잡음 성능을 향상시키기 위해 회로에서 발생하는 광대역 잡음을 개선해야 한다고 주장해 왔다. 그러나 본 연구에서는 세포와 전극 사이 전기적 네트워크를 분석하여 90% 이상의 잡음이 실링 저항(sealing resistance)에서 발생하는 것을 확인하였다. 또한 광대역 잡음 상쇄 기법을 사용하여 실링 저항에서 발생하는 잡음을 효과적으로 줄이고 측정되는 신호의 신뢰도가 높아지도록 하였다. 이를 통해 $10 \mu m$ 간격 고밀도 조건과 100,000 전극의 고집적 조건에서 전 프레임 측정이 효과적으로 이루어질 수 있었다. 광대역 잡음 상쇄 기법을 사용하지 않은 조건과 비교하여 실링 저항에서 발생하는 잡음이 85% 감소하였으며 입력 등가 잡음은 $80 \mu Vrms$ 에서 $18 \mu Vrms$로 감소하였다. 신호 대 잡음 비가 전체적으로 크게 향상되었으며, 기존 구조에서는 잡음에 가려 볼 수 없었던 약한 실링 조건에서의 신경 신호도 획득할 수 있었다.
