The dentate gyrus (DG) is known to be critical for detecting changes in environments, however the specific roles of main excitatory cell types of DG: granule cells (GC) and mossy cells (MC) are still unknown. Here we designed reusable optoelectronic probe for chronic single unit recording and optogenetic manipulations. By implementing optogenetic labeling using this tool, I was able to identify electrophysiological features of GCs and MCs. I observe that fixing a new object, while mice running on the familiar belt, induces a distinct GC and MC response, with maximal alteration near the new object. In general, MCs response is stronger and peaks earlier than GCs. These results suggest that MC spread information about change to GC network thus helping for pattern separation.
치상회는 환경 속에서 변화를 감지하는데 중요한 역할을 하는 것으로 알려져 있지만, 치상회의 주요 흥분성 세포인 과립세포와 이끼세포가 어떻게 기여하는지에 대해서는 알려지지 않았다. 이 연구는 세포의장기레코딩을하기위한광전극을재사용할수있는툴을 설계하고,이툴을이용해광유전학 적 표지를 이용한 과립세포와 이끼세포의 특성을 확인하였다. 쥐가 익숙한 트래드밀 벨트 위를 달리는 동안 새로운 물체를 넣어주었을 때 과립세포와 이끼세포가 새로운 물체 근처에서 세포 활성이 변화하는 것을 보았다. 일반적으로 이끼세포의 경우 과립세포보다 강하고 빠르게 세포 활성이 변화하였다. 이 결과는 이끼세포가 변화를 감지하여 과립세포의 네트워크에 정보를 전달하여 패턴 분리에 기여하는 것을 시사한다.
