To develop the treatment to reduce muscle soreness, we tried to investigate the effect of hypoxia and cyclic stretch on the recovery of oxidative damaged skeletal muscle cells. C2C12 myoblast cells were differentiated and grown in the microfluidic device then induced oxidative stress by 1mM hydrogen peroxide. After the damage-inducing, damaged myotubes were repaired in four different environments, with and without cyclic stretch in normoxia (21% O2) and hypoxia (6% O2) to figure out the effect of hypoxia and cyclic stretch on recovery through comparing each condition. Reactive oxygen species, 8-hydroxydeoxyguanosine, and myosin heavy chain were chosen as recovery indicators. It was confirmed that hypoxia and cyclic stretch facilitated the recovery of muscle cells and the recovery was best performed in hypoxia with the cyclic stretch condition. To investigate the mechanism of the hypoxia and stretch intervened recovery, Hypoxia inducible factor -1$\alpha$ (HIF-1$\alpha$), Peroxiredoxin 2, Glutathione peroxidase 1, and Catalase were selected as candidates for the key factor of mechanism. HIF-1$\alpha$ and antioxidants seemed strongly related to enhanced recovery by hypoxia and cyclic stretch, and HIF-1$\alpha$ was speculated to play the most important role. By integrating the whole results, we were able to suggest the stretch and hypoxia triggered the pathway of cellular recovery.
근육통을 줄이기 위한 치료법을 개발하기 위해 산화적 손상을 입은 골격근의 회복에 대한 저산소증과 주기적 장력이 미치는 영향을 연구했다. 근육 모세포를 미세유체 칩에서 배양시킨 후 과산화수소에 의한 산화적 손상을 유발하였다. 이후 손상된 근섬유는 일반적인 대기 (21% 산소)와 저산소 대기 환경 (6% 산소)에서 주기적 장력의 유무에 따른 총 4가지 환경에서 각각 회복되어 각 조건에 따른 회복 정도를 비교함으로써 저산소 환경과 주기적 장력이 회복에 미치는 영향을 분석했다. 분석에는 활성산소, 8-하이드록시 데옥시구아노신, 미오신 중쇄와 근섬유의 두께가 지표로 사용되었다. 저산소증 및 주기적 장력이 근육 세포의 회복을 촉진하고 두 가지 조건 모두 동시에 가해줬을 때 가장 회복이 잘 되었음을 확인했다. 저산소증과 주기적 장력에 의한 회복 기작을 조사하기 위해 저산소유도 인자-1$\alpha$, 퍼옥시리독신 2, 글루타싸이온 퍼옥시더레이즈 1, 그리고 카탈라제를 해당 기작의 주요 인자로 추정하였다. 저산소유도 인자-1$\alpha$및 항산화제는 저산소증 및 주기적 장력에 의한 회복 회복과 밀접한 관련이 있는 것으로 보였으며, 그 중에서도 저산소유도인자-1$\alpha$가 가장 중요한 역할을 하는 것으로 보인다. 본 연구를 통해 우리는 스트레치 및 저산소증이 세포 회복 경로를 유발한다는 것을 제안할 수 있었다.
