Intracellular antibodies, such as nanobodies and single-chain variable fragments (scFv), have become powerful tools for imaging, modulating, and neutralizing endogenous target proteins. Although a variety of antibody engineering techniques have been developed, an optogenetic tool for activating intracellular antibody to precisely control its target protein has not been reported. In chapter 1, I describe an optogenetically activatable intracellular antibody (Optobody) consisting of split antibody fragments and blue light-mediated heterodimerization domains. Blue light stimulation activates the optobody, inducing it to capture its target protein and subsequently inhibit the target. In chapter 2, I expanded this optobody platform by generating various optobodies from previously developed intracellular antibodies. In chapter 3, I demonstrated that photoactivation of a gelsolin (GSN) optobody and $\beta$2 adrenergic receptor ($\beta$2AR) optobody shut down endogenous GSN activity and $\beta$2AR signaling, respectively. Applying the novel optogenetic platform to the broad pool of available intracellular antibodies will facilitate optogenetic manipulation of various endogenous proteins and may provide a basis for designing potential inducible drugs.
나노바디나 scFv 와 같은 세포 내 항체는 내생단백질을 이미징하거나 그 기능을 조절하는 기술로 각광받고 있다. 많은 항체 공학 기술이 개발되고 있지만, 세포 내 항체의 활성을 시공간적으로 조절하는 기술은 아직 보고되지 않았다. 본 학위논문에서는 빛으로 항체의 활성을 유도하는 광활성 항체 기술 (옵토바디)의 개발 및 적용에 관하여 다루고자 한다. 제 1장에서는 기술을 개발하고 그 기술적 특성을 규명한다. 본 기술은 둘로 쪼개진 비활성 항체를 청색광을 통해 재결합하여 항체 활성을 유도시킨다. 제 2장에서는 본 기술의 범용성을 보여주기 위해 기존에 개발된 다양한 항체 조각을 이용하여 내생단백질을 인지하는 옵토바디들을 제작 및 소개한다. 제 3장에서는 광활성 항체 기술을 이용해 내생단백질의 기능을 저해함으로서 본 기술이 단백질 기능 억제제로 사용가능함을 보여준다. 원하는 위치와 시간대에 항체의 활성을 유도하는 본 기술은 단백질 기능을 밝히는 연구분야에 활용성이 크며, 새로운 개념의 잠재적 의약품으로 발전될 가능성이 있다.
