Since the increase of average human lifespan and population aging, the importance of early diagnostics has been raised and the market of in vitro diagnostic (IVD) has been increased consistently in recent 10 years. We herein described novel detection methods for biomolecule by utilizing the conformational change of hairpin probe induced by target molecule. In this study, we devised a detection system for target protein and activity of ribonuclease H (RNase H) based on the DNA polymerase activity, which is regulated by the combination of hairpin structured DNA aptamer and target recognition strand. Furthermore, by introducing the self-priming re-action of hairpin probe to isothermal nucleic acid amplification, we successfully simplified the reaction mecha-nism that is commonly complicated in isothermal amplification methods and developed highly sensitive detec-tion system for target nucleic acid. Since the devised systems are available for quantitative analysis of target molecules in a real-time manner, it is expected that the developed system would be a potential sensing tool in the field of IVD testing.
평균수명 연장과 고령화 사회로의 진입으로 인해, 질병의 조기진단의 중요성이 대두됨에 따라, 체외진단 시장은 지난 10년 간 지속적인 성장세를 보였으며, 향후 더욱 급격한 성장이 전망된다. 본 연구에서는, 표적 물질에 의해 야기되는 헤이핀 프로브의 구조 변화를 이용하여 새로운 생체물질 검출 기술을 개발하였다. 먼저, DNA 중합효소와 특이적으로 상호작용하여 효소 활성을 억제하는 헤어핀 구조의 DNA 압타머를 기반으로 표적 단백질 및 리보뉴클리아제 H의 활성을 DNA 중합 반응의 구동 여부를 통해 검출 할 수 있는 기술을 개발하였다. 뿐만 아니라, 헤어핀 프로브의 self-priming 반응을 접목한 등온 핵산 증폭 반응을 개발하여, 등온 핵산 증폭 반응에 수반되는 복잡한 반응 메커니즘을 단순화하였으며, 이를 기반으로 고민감도의 표적 핵산 검출 기술을 개발하였다. 본 연구에서 개발된 헤어핀 프로브의 구조 변화를 기반으로 한 생체물질 검출 기술은 표적 물질의 실시간 정량 분석이 가능함에 따라, 향후 체외 진단 분야를 선도하는 핵심 기술로 자리잡을 수 있을 것으로 기대된다.
