Heterotrophic cultivation has notable benefits, including high cell density and the ability to produce biomass in a consistent and predictable amount and the relatively low risk of contamination. Heterotrophic cultivation of Chlorella sp. HS2 was optimized in this study. First, various sources of carbon and nitrogen were selected in the photobiobox. Next, Plackett-Burman Design (PBD) was used to check the effect of each nutrient on cell growth. And the concentration of the major nutrients (glucose, nitrate, and phosphate) was optimized through the Response Surface Methodology (RSM) using the Central Composite Design (CCD). The model verification results through flask cultivation showed that the modified BG11 medium improved the total cell production by 3-fold from 5.85 to 18.13 g $L^{-1}$ compared to the normal BG11 medium. Expanding the incubation scale to a 5-L fermenter greatly improved biomass concentration of 35.3 ± 0.6 g $L^{-1}$ due to more efficient cultivation. Compared with other Chlorella spp., the biomass concentration was significantly higher, but the lipid content was low. To overcome these problems, I tried to solve them through fed-batch cultivation. The BG11 medium was also used to do fed-batch cultivation by starting incubation and adding the optimized BG11 medium. As a result, the biomass concentration was increased from 32 g $L^{-1}$ to 50 g $L^{-1}$. In the case of lipid content, it also increased sharply from 26% to 40%. But there were some weak parts here, too. In addition to glucose, there was a buildup of other nutrients. So I tried again by adding nutrients only the required amount of biomass to the target biomass concentration. And the dissolved oxygen was also found to have affected cell growth by falling from days 2-3 to zero. In order to solve this problem, I want to gradually increase RPM. As a result, biomass grew beyond 50 g $L^{-1}$ in just 5 days, and existing biomass production doubled. Dissolved oxygen was kept above 20%. Glucose, as it had been cultured earlier, maintained a good 20-40 g $L^{-1}$ and nitrate remained after 4 days. On the 8 day, when lipid production was the highest, lipid production was 2.29 g $L^{-1}$$d^{-1}$, an improvement from 1.99 g $L^{-1}$$d^{-1}$, the lipid productivity of the fed-batch cultivation initially conducted. However, lipid content decreased by about 7.6% points from 39.8% to 32.2%. In order to increase lipid production, nitrate was given in half to try the fed-batch cultivation with nitrogen depletion stress again. As a result, nitrate was depleted in 3 days and lipid production edged up to 2.31 g $L^{-1}$$d^{-1}$ on the 8 days when lipid production was the highest. Here, biomass grew rapidly to 47 g $L^{-1}$ in just 3 days. The transcriptome analysis was attempted to further interpret the difference in lipid accumulation process seen in the fed-batch of the nitrogen replete and depleted conditions. As a result, we successfully increased total lipid productivity in the Fed-batch experiment. Then, we predicted and identified phenotype of N-limited cultivation by confirming expression level of N-limited mRNA. We tried transcriptome analysis to explain the special regulatory mechanism about N-limited cultivation. Then, quantified gene expression between N-sufficient and N-limited cultivation was compared, and gene ontology was conducted. Based on these data, we predicted specific genes that could be regulated by Chlorella sp. HS2 under specific culture conditions. These results suggest new possibilities of upgraded cultivation process engineering of Chlorella sp. HS2 for the production of valuable bio-product and biofuels.
미세조류의 종속 영양 배양은 높은 세포밀도로 배양이 가능하다. 게다가 일관되고 예측 가능한 양으로 바이오매스의 생산이 가능하며 오염에 대한 위험이 상대적으로 적어 단일 균주만 배양 가능 등 많은 주목할 만한 이점을 가지고 있다. 이번 연구에서 사용된 균주는 클로렐라 HS2로 기존에는 광합성 배양만을 시도했던 미세조류이다. 그래서 우선 종속 영양 배양을 위해 배지의 최적화를 시도하였다. 첫 번째로 포토바이오박스에서 다양한 탄소 및 질소 공급원을 선별하였다. 다음으로 Plackett-Burman Design (PBD)를 이용해 배지내 각각의 영양분들이 세포 성장에 미치는 영향을 확인하고, 주요 영양소(포도당, 질산염, 인산염)의 농도는 중앙 복합 설계 (CCD)로 반응 표면 방법론 (RSM)을 통해 최적화되었다. 플라스크 배양을 통한 모델 검증 결과, 최적화된 최적화된 BG11 배지는 최적화되지 않은 BG11 배지와 비교하여 5.85에서 18.13 g $L^{-1}$로 세포 총 생산량이 3배 향상되는 결과를 보였다. 배양 규모를 5-L 발효기로 확대하면 보다 효율적인 배양으로 인해 35.3 ± 0.6 g $L^{-1}$의 바이오매스 농도가 크게 개선되었다. 다른 클로렐라와의 비교에도 상당히 높은 바이오매스 농도를 배양하였지만, 지질함량은 낮게 나왔다. 이러한 문제를 극복하기 위해 유가식 배양을 통해 해결하고자 하였다. 유가식 배양에서도 최적화된 BG11배지를 사용해서 배양을 시작하고 최적화된 BG11 배지를 추가적으로 넣어주는 방식으로 배양하였다. 그 결과 기존 회분식 배양에서 바이오매스 농도가 32 g $L^{-1}$인 것에 비해 10일 만에 50 g $L^{-1}$까지 올라간 것을 확인하였다. 지질 함량의 경우에도 26%에서 40%로 대폭 상승했다. 하지만 여기서도 약간의 보완할 부분들이 있었다. 우선 글루코스 이외에도 다른 영양분들이 추가로 넣어주면서 쌓이는 현상이 발생하였다. 그래서 목표하는 바이오매스 농도까지 필요한 정도만 처음에 넣어주는 방법으로 다시 시도하였다. 그리고 용존 산소량의 경우도 2-3일차에서 0이 되어 세포 성장에 영향을 미쳤을 것으로 확인된다. 이러한 점을 해결하기 위해 RPM을 점차적으로 올려줘서 해결하고자 한다. 그 결과 5일만에 바이오매스는 50 g $L^{-1}$를 넘어서 성장하였고 기존 바이오매스 생산량이 2배 증가하였다. 산소는 20% 이상으로 유지를 하였다. 앞서 배양했던 것처럼 글루코스는 20-40 g $L^{-1}$를 잘 유지해주었고 질소염은 4일차 이후로 고갈이 되지 않고 남았다. 지질 생산량이 가장 높았던 8일차에 지질 생산량은 2.29 g $L^{-1}$$d^{-1}$로 처음에 진행한 유가식 배양의 지질 생산량인 1.99 g $L^{-1}$$d^{-1}$보다 향상되었다. 하지만 지질 함량은 39.8%에서 32.2%로 약 7.6% 포인트 감소하였다. 지질 생산량을 높이기 위해 질산염을 절반으로 주어 질산염 고갈 스트레스가 있는 유가식 배양을 다시 시도하였다. 그 결과 질산염은 3일 만에 고갈이 되었고 지질 생산량이 가장 많았던 8일차에 지질생산량이 2.31 g $L^{-1}$$d^{-1}$로 소폭 상승되었다. 여기서 바이오매스가 3일만에 47 g $L^{-1}$까지 빠른 성장을 보였다. 앞의 질산염의 고갈이 되지않은 조건과 고갈된 조건의 유가식 배양에서 보이는 지질 축적 현상에 차이를 자세하게 해석하기 위해 전사체 해석을 시도하였다. 그 결과, 유가식 배양 실험에서 총 지질 생산성을 성공적으로 증가시켰다. 그런 다음 질소 제한 mRNA의 표현 수준을 확인하여 질소 제한 배양의 표현형을 예측하고 식별하였다. 우리는 질소 제한 배양에 관한 메커니즘을 설명하기 위해 transcriptome 분석을 시도했다. 그 다음, 질소 충분 조건과 질소 제한 배양 사이의 정량화된 유전자 발현을 비교하고, gene ontology를 실시했다. 이러한 데이터를 바탕으로 클로렐라에 의해 조절될 수 있는 특정 유전자를 예측했다. 고부가가치의 바이오 제품과 바이오 연료 생산을 위한 특정한 배양 조건 하에서 이러한 결과는 클로렐라의 개량된 배양 공정의 새로운 가능성을 시사한다.
