Toll-like receptors(TLRs) play key roles in the initiation of the innate immune system by recognition of conserved foreign molecules called pathogen-associated molecular patterns. TLR 3 is located in endosomes and it recognizes virus infection by sensing dsRNA which is produced during virus replication. Especially, TLR3 is related to CNS virus infection. However, the activation mechanism and downstream signaling pathways are unclear because of considering only ectodomain and lack of complete structural information. In this research, we focused on expression and purification of full-length of TLR3 which is essential for revealing activation mechanisms by structural studies. To provided correct post-translation modification and lipid environments, TLR3 was expressed by the mammalian expression system – bacmam. Solubilization detergent that is one of the most important for membrane protein stability could be determined by FSEC. Finally, the sample quality was check by negative-stained TEM.
톨 유사 수용체는 박테리아나 바이러스의 패턴 분자 인식을 통해 외래 물질 침입을 인식하여 선천성 면역을 시작하는데 중요한 역할을 한다. 그 중에서 톨 유사 수용체 3은 엔도좀에 위치하여 바이러스 복제 과정에서 생성되는 dsRNA를 인식해 바이러스 감염을 감지한다. 바이러스감염 중 특히 중추신경 감염과 큰 관련이 있다. 이러한 중요성에도 불과하고 Ectodomain에 국한된 구조연구로 인해 톨 유사 수용체 3의 활성 메커니즘과 신호전달은 불분명하다. 본 연구는 톨 유사 수용체 3의 활성 메커니즘을 밝히기 위한 단백질 구조연구에 있어 필수적인 전체 길이의 톨 유사 수용체 3 대량 발현 및 정제 방법을 연구하였다. 올바른 번역 후 변형 및 지질환경 제공을 위해 바큘로바이러스를 이용한 포유동물 세포에서 발현 시스템을 통해 발현하였고 이후 단백질을 세포막으로부터 가용화하기 위한 조건으로 여러 계면활성제 조건을 테스트하였다. 최종적으로 샘플의 상태를 음성염색법과 전자현미경을 통해서 확인하였다.
