Many intracellular proteins with great potential as biopharmaceutical drugs have been identified; however, numerous challenges associated with intracellular protein delivery have yet to be solved. Recent studies have sought to use exosomes as a new method for the in vivo delivery of various cargos to specific target tissues by systemic injection. Soluble proteins can be loaded into the exosome via controlled, reversible, light-induced protein-protein interactions (PPI) named "exosomes for protein loading via optically reversible protein-protein interaction” (EXPLOR). I utilized opto-genetically engineered exosome system to load β-glucocerebrosidase (GBA), which is an enzyme deficient in Gaucher disease patients, into newly generated exosomes. In the present study, I demonstrated the intracellular delivery of beta-glucocerebrosidase as functional proteins from the exosomes to the target cells, indicating the potential of EXPLOR for treatment of Gaucher disease.
세포 내에 존재하는 단백질 중에는 바이오의약으로 활용될 가능성이 높은 단백질들이 많이 존재한다. 그러나 이들을 다른 세포 안으로 전달하여 약물로써 사용하는 것에는 아직까지 많은 한계가 있다. 최근에는 엑소솜을 활용하여 다양한 약물들을 그 안에 탑재하여 생체 내에 특정한 조직으로 전달하는 전달 방법이 주목을 받고 있다. 수용성 단백질들은 엑소솜 안에 광유전학적 기법을 통해 빛으로 유도된 단백질-단백질 상호작용으로 인하여 탑재가 될 수 있다. 본 연구에서는 이 기술을 활용하여 광유전학적 기법으로써 고셰병 환자에서 결핍을 보이는 효소인 글루코세레브로시다제를 탑재한 엑소솜을 제작하였다. 이러한 엑소솜 안에는 효소의 활성도를 가지고 있는 효소를 탑재하고 있으며, 또한 이를 고셰병 환자에서 유래된 세포에 처리를 해줌으로써 환자 세포에서 결핍을 보였던 효소의 활성을 회복시킨 것을 확인하였다. 본 연구를 통해 엑소솜-기반 효소 전달 치료가 효소 결핍증 환자 치료에 활용될 수 있는 가능성을 시사하였다.
