We herein developed a novel target RNA detection method called Nicking/Extension chain reaction System-Based isothermal Amplification (NESBA). In principle, the presence of target RNA induces formation of double-stranded (ds) DNA containing nicking sites and T7 promoter sequence which are utilized for nicking/extension chain reaction- and transcription-mediated isothermal amplification, respectively. As a result, RNA amplicons bind to the molecular beacons (MBs) leading to highly enhanced fluorescent signal. Based on this novel strategy, we successfully detected target genomic RNA (gRNA) derived from influenza A (H3N2) virus down to 3.6 ag/$\mu$L with high specificity over other gRNAs from different subtypes of influenza A virus.
본 학위논문에서는 핵산의 절단 및 중합연쇄반응 시스템 기반 등온 증폭 기술을 이용한 새로운 표적 RNA 검출 기술을 개발하였다. 본 기술은 표적 RNA가 존재할 경우, 등온 증폭 반응에 사용되는 절단 효소 인식 서열과 T7 프로모터 서열을 동시에 포함한 이중 가닥 DNA를 생성한다. DNA로부터 일어나는 절단 및 중합연쇄반응과 전사 반응 기반의 등온 증폭 반응에 의해 최종 생성된 RNA 산물들은 형광물질이 수식된 분자 비콘과 반응하여 높은 형광 신호를 발생시킨다. 상기 시스템을 기반으로, H3N2 인플루엔자 바이러스의 표적 유전체 RNA를 3.6 ag/$\mu$L의 낮은 검출 한계로 검출하였을 뿐만 아니라 H3N2 이외의 다른 종류의 인플루엔자 바이러스 검출을 통해 높은 선택도를 확보하여, 성공적으로 본 기술의 실용성을 증명하였다.
