Recently, phase separation is known to play a role in the formation of various membraneless cellular organelles. These organelles are suggested as supramolecular assemblies of proteins and RNA molecules, that accumulate its comprising element and other biomolecules. This localization causes enrichment of the local concentration in phase-separated droplets. However, the detailed principle in increasing local concentration effects on biological kinetics is undiscovered. This study suggests behavior aspects of localized biomolecules in phase-separated droplets by a simplified model system. Using proline-rich motifs (PRM) and the SRC homology 3 (SH3) domain interaction, metal-induced protein phase separation scaffold was constructed as a model system. In the PRM-SH3-His (PSH) model scaffold, I develop interactable client proteins adjusted by the number of PRM. As a stable reaction product, isopeptide bond formation reaction was adopted as a target reaction. To follow reaction, fluorescent protein and dye were used. As a result, more interaction motifs cause higher local concentration. However, the overly high local concentration of scaffold protein and strong interaction like a nickel with 6x histidine tag are restricted molecular movement which comes up as decline of reactivity.
최근 들어 세포 내 막이 없는 세포소기관들에 대한 연구가 활발히 진행되고 있다. 하지만 이들의 형성 원리와 역할에 대한 세부적인 연구는 기초 단계에 머물러 있다. 이러한 세포소기관들이 상분리 현상을 통하여 이루어지고 내부에 다른 여러 생체 분자들을 축적하는 사실만이 알려져 있는 시점이다. 따라서 본 연구 논문에서는 생체 분자를 이용하여 구상한 간략화된 모델계를 통하여 상분리 된 구획 내부의 농도에 초점을 맞추어 다른 생체 분자의 농축과 이에 따른 이소펩타이드 결합의 반응성에 대하여 연구를 진행하였다. 그 결과 상분리 구획 내 농도와 분자간의 상호작용 세기에 따라 반응성이 달라짐을 관찰할 수 있었다.
