We herein describe a simple, enzyme-free, and label-free strategy for the detection of miRNA based on target-triggered catalytic hairpin assembly (CHA) and fluorescence enhancement of DNA-silver nanoclusters (DNA-AgNCs). This strategy employs two hairpin probes that are rationally designed to contain AgNCs and guanine (G)-rich DNA sequence, respectively. In principle, the two hairpin probes, which exist independently, form a hybridized complex in the presence of target miRNA through CHA reaction, thereby inducing strong fluorescence signal of AgNCs by the presence of G-rich sequence placed in close proximity. Importantly, target miRNA is released after the first CHA reaction, which promotes another CHA reaction, consequently leading to the significantly enhanced fluorescence signal of AgNCs. Based on this one-step, homogeneous, and isothermal amplification method, we successfully detected the target miRNA with high selectivity and sensitivity. In addition, the practical applicability of this strategy was also demonstrated by analyzing the target miRNA in human serum. Moreover, the effect of hairpin stem and toehold length was assessed thoroughly, providing the improved understanding of the kinetics of CHA reactions.
본 연구에서는 헤어핀 연쇄 조립과 다수 구아닌에 의한 DNA 은나노입자의 형광 증가를 이용하여 microRNA를 검출할 수 있는 무효소, 비표지 방법을 개발하였다. 본 기술은 은나노입자와 다수 구아닌을 각각 포함하는 두 개의 헤어핀 DNA 프로브를 이용한다. 두 개의 헤어핀 프로브는 서로 독립적으로 존재하지만 표적 microRNA의 존재 하에서는 헤어핀 연쇄 조립을 통하여 혼성화하여 복합체를 이루며, 이는 은나노입자에 다수 구아닌을 근접시켜 강한 형광 신호를 발생시킨다. 표적 microRNA 는 헤어핀 연쇄 조립 반응 후에 방출되어 새로운 헤어핀 프로브와 다시 결합을 이룸으로써 또 다른 헤어핀 연쇄 조립 반응을 일으키며 은나노입자의 형광 증폭을 초래한다. 본 기술은 1 단계, 균질, 등온 반응이라는 장점을 가지고 있으며 이 기술을 통하여 표적 mircroRNA를 민감하고 선택적으로 검출할 수 있다. 또한 인간 혈청 안의 microRNA 를 검출함으로써 본 기술의 실용성이 입증되었다. 또한 본 연구에서는 헤어핀 프로브의 구조가 헤어핀 연쇄 조립에 미치는 영향이 철저히 분석되어 헤어핀 연쇄 조립법의 역학적 이해를 도울 수 있을 것으로 기대된다.
