Many organisms have evolved to protect themselves from external stress by particular defensive sys-tem such as eggshells and bacterial endospore. Previous research has examined the formation of artificial shells which have resistance to adverse change. Living cells in artificial shells can behave depending on their innate character because artificial shells play a role of protector. There are several methods widely used to encapsulate cells such as Layer-by-Layer (LbL) techniques, microfluidic techniques, and mineralization. However, they have mainly focused on the single cell encapsulation. Otherwise, cells couldn’t move freely because they were immobilized in case of research about multicell encapsulation, Accordingly, it is necessary to encapsulate several living cells with free space in which individual cells move freely.
In this study, we propose synthesis method of micro polyelectrolyte capsules encasing multiple living cells using both LbL assembly and mineralization. The combination of these two techniques enables living cells to be enveloped with sufficient free space. Through mineralization of $CaCO_3$, several living cells could be entrapped in $CaCO_3$. Negatively charged cell surface acted as the site of nucleation and growth of $CaCO_3$. Layer-by-Layer assembly performed onto $CaCO_3$ as sacrificial templates. The dissolution of $CaCO_3$ core resulted in the synthesis of polyelectrolyte shells containing several living cells with sufficient empty spaces.
As a result, cell embedded $CaCO_3$ and polymer capsules encasing living cells were successfully synthesized. Through control of the concentration of $CaCO_3$ source and microalgae, the number of algae in the polymer shell and free space could be adjusted. In addition, different cells could be successfully encapsulated. Moreover, these capsules could be easily functionalized to respond to specific external stimuli by using nanomaterials. Consequently, cell could behave and prolife in the newly suggested capsules. These capsules also act as microenvironment to observe cell behaviors.
많은 생물들은 외부 환경 변화로부터 스스로를 보호하기 위한 체계를 가지고 있다. 달걀, 조개, 달팽이 껍질 등을 그러한 예로 들 수 있다. 박테리아의 경우 영양 결핍, 고온 등의 환경에서 자신을 보호하기 위해 포자를 형성한다. 이러한 점에서 착안하여 세포에 인공적인 껍질을 쌈으로써 세포를 보호하고자 하는 세포 캡슐화에 대한 연구가 활발히 진행되고 있다. 세포에 인공적인 껍질을 감싸게 되면 세포의 생존성이 증가할 뿐 아니라 외부 환경으로부터 영향을 적게 받고 거동 할 수 있게 된다. 미세유체기술, 광물화, Layer-by-Layer 법 등을 이용하여 세포 캡슐화 연구가 진행되어 왔는데, 대부분 단일 세포에 적용되거나 세포가 하이드로겔과 같은 매트릭스에 고정되어 움직일 수 없는 상태였다. 그렇기에 여러 개의 세포를 동시에 캡슐화 하면서도 자유 공간을 두어 세포가 자유롭게 거동할 수 있도록 하는 세포 캡슐화 방법이 필요한 상황이다.
본 연구에서는 광물화와 Layer-by-Layer 법 두가지 방법을 사용하여 마이크로 사이즈의 여러 개의 세포를 감싸는 고분자 캡슐을 합성하였다. 음전하를 띄는 세포의 표면은 결정 성장의 자리가 되어 여러 개의 세포를 감싸며 탄산 칼슘 결정이 자랄 수 있도록 해주었다. 이 때 탄산칼슘 결정의 생성 반응 시간 및 반응물의 농도를 조절해줌으로써 두가지 상의 세포를 감싼 탄산칼슘이 형성 될 수 있었다. 또한 탄산칼슘 결정을 생성하는 반응물의 농도 혹은 세포의 농도에 따라서 결정에 갇히는 세포의 수나 결정의 크기도 조절 될 수 있었다. 탄산칼슘 결정을 템플릿으로 하여 Layer-by-Layer 법을 통해 고분자 전해질을 흡착시켰고, 결과적으로 탄산 칼슘을 녹여 냄으로써 고분자 캡슐이 합성되었다. 고분자 캡슐이 전하를 띄기 때문에 나노 입자로도 쉽게 코팅되었고, 이는 캡슐이 빛이나 자기장 등 특정한 자극에 반응할 수 있도록 적용될 수 있다. 고분자 캡슐은 여러 개의 세포들이 충분한 자유공간을 가지면서 움직이고 분화할 수 있도록 합성되어 시간이 지남에 따라 캡슐 속 세포가 자라 캡슐을 가득 채우는 것을 확인할 수 있었다. 즉, 캡슐이 세포가 자라고 관찰 할 수 있는 미세 환경으로서의 역할 또한 할 수 있다는 것을 의미한다.
