Ginseng is a medicinal herb that requires cultivation under shade conditions, typically for 6 yr, before harvesting. The principal components of ginseng are ginsenosides, glycosylated tetracyclic terpenes. Ginseno-sides are classified into two groups, protopanaxadiol (PPD) and protopanaxatriol (PPT), based on their hy-droxylation patterns, and further diverge to diverse ginsenosides through differential glycosylation. Three early enzymes, Dammarenediol-II synthase (PgDS) and two P450 enzymes, protopanaxadiol synthase (PgPPDS) and protopanaxatriol synthase (PgPPTS), have been reported, but glycosyltransferases that are necessary to synthesize specific ginsenosides have not yet been fully identified. To discover glycosyltransferases responsible for ginsenoside biosynthesis, we sequenced and assembled Panax ginseng transcriptome de novo and characterized four UDP-glycosyltransferases (PgUGTs): PgUGT74AE2, PgUGT94Q2, PgUGT71A27 and GpUGT23. PgUGT74AE2 transfers a glucose moiety from UDP-glucose (UDP-Glc) to the C3 hydroxyl groups of PPD and compound K to form Rh2 and F2, respectively, whereas PgUGT94Q2 transfers a glucose moiety from UDP-Glc to Rh2 and F2 to form Rg3 and Rd, respectively. PgUGT71A27 introduced glucose moiety to C20 hydroxyl group of PPD, Rh2, Rg3 and PPT and produced CK, F2, Rd and F1, respectively. GpUGT23 introduced glucose moiety to C20 position glucose of CK, F2, Rd and F1 and produced Gypenoside XLLV, gypenoside XVII, ginsenoside Rd and notoginsenoside U, respectively. Introduction of the combination of four UGTs into yeast together with PgDS and PgPPDS resulted in the de novo production of Rg3, F2, F1, Rd, CK and gypenoside XVII. Our results indicate that these four UGTs are key enzymes for the synthesis of ginsenosides and provide a method for producing specific ginsenosides through yeast fermentation
진세노사이드는 인삼의 약리효과를 나타내는 주성분으로 구조적인 차이에 의하여 Pro-topanaxadiol (PPD)와 Protopanaxatriol (PPT) 두 개의 그룹으로 나눌수 있다. 또한, 전체 진세노사이드안에 포함된 양에 따라 많은 양이 포함되어 있는 메이저 진세노사이드와 적은 양만이 존재하는 마이너 진세노사이드로 나뉜다. 메이저 진세노사이드는 약한 약리효과를 보이며 진세노사이드 Rd, Rb1, Rb2, Rb3, Rd, Rg1등이 있고, 마이너 진세노사이드는 강한 약리효과를 가지고 있고, Rh2, Rg3, F2, F1, Rh1 등이 있다. 기존에는 진세노사이드를 인삼에서 추출하여 생산을 하고 있으나, 낮은 생산성과 여러 물질의 혼합물로 생산이 된다는 문제점이 존재한다.
단일물질을 대량생산하기 위하여 미생물에 진세노사이드 대사회로를 도입을 하여 생산균주를 개발하고 미생물을 이용하여 생산하는 연구를 시작하였다. 이 연구 또한 문제가 존재 하고 있었는데, 구축하고자 하는 대사회로에 대한 유전정보가 밝혀져 있지 않아 대사회로를 구축을 하려면 유전정보를 밝히는 연구가 선행이 되어야 했다. 인삼의 genome sequencing이 완료가 되지 않아, 직접 sequence분석을 하였는데, 이를 위하여 MeJA를 처리한 인삼 잎과 뿌리를 이용하여 Next generation sequenceing method를 이용하여 RNA transcriptome analysis를 하여 MeJA에 의해 과발현된 유전자 시퀀스를 분석하여 진세노사이드 합성에 관련된 유전자들을 선별하고 활성분석을 통하여 실제 진세노사이드 합성 기능이 있는 유전자들을 확보하였다. 진세노사이드 합성에 있어 핵심 유전자인 UDP-glycosyltransferase (UGT)에 대한 정보는 알려진 바가 없었는데, 총 4종류의 UGT를 확보하였고, in vitro 실험 결과 UGT들의 조합을 통해 다양한 진세노사이드를 만들수 있음을 확인하였다.
다음으로, 확보한 UGT들과 기존에 알려진 유전자들을 이용해 효모인 S. cerevisiae에 진세노사이드 합성 경로를 도입하여 균주를 개발하였다. 먼저 진세노사이드 합성 경로와 com-petition을 이루는 Ergosterol synthesis pathway를 down regulation 할 수 있는 S. cerevisiae MET3p-ERG7 균주를 만들었고, 이 균주에 Dammarenediol-II synthase, Protopanaxadiol synthase, Protopanaxatriol synthase, PgUGT74AE2, PgUGT94Q2, PgUGT71A27 과 GpUGT23들을 목적 물질에 맞춰 도입하여 ginsenoside Rg3, F1, F2, Rd, Compound K, gype-noside XVII을 각각 만들 수 있는 6종류의 균주를 만들어 Fed-batch fermentation을 하여 실제로 제작한 균주들을 이용해 각각의 진세노사이드들을 생산하는데 성공하였다.
생산량을 높이기 위하여 아직 해결 해야 하는 부분이 남아 있지만, 이 연구 결과를 통하여 인삼의 기능성 물질인 진세노사이드를 식물이 아닌 미생물을 이용하여 필요한 단일 물질만을 생산 할 수 있다는 것을 증명하였고, 진세노사이를 이용한 대양한 산업 발달의 초석을 다졌다고 생각한다.
