Graphene quantum dots (GQDs) are expected to be used in bio-fields due to unique optical properties and low toxicity. However, the conventional synthesis methods have disadvantage such as poor quality of GQD and complicated production process. Therefore, by applying the silicon carbide (SiC), a well-known material in synthesis of epitaxial graphene, GQD was fabricated by heating SiC in a medium vacuum. GQD solution was prepared by detached GQDs from SiC. As prepared GQD was composed of high quality graphene and had a uniform size and thickness. The PL signal of pristine GQD-ethanol solution without any treatment was too weak for using bio-imaging field. However, when additional sonication process for 30minutes was performed to as prepared GQD solution, the intensity of PL was dramatically increased. The surface of the GQD was found to be oxidized after an additional sonication process applied to the GQD through XPS analysis. It was presumed that the surface passivation was the origin of photoluminescence of GQD solution. GQD showed low cytotoxicity and it was observed that cells were maintained in a normal state even after GQD was injected into the cells in the cytotoxicity tests. Thus, it is expected that GQD fabricated by annealed SiC will be used in bio-field at future.
그래핀 양자점(GQD)은 낮은 독성과 우수한 발광 특성으로 인해 바이오 분야에서 다양하게 이용가능하다. 그러나 기존의 합성 방법은 합성된 GQD의 순도가 낮고 제작과정이 복잡하다는 단점이 있다. 따라서 실리콘 카바이드(SiC)를 고온에서 가열하면 에피택셜 그래핀이 합성되는 사실을 응용하여 중 진공에서 SiC를 가열하여 GQD를 합성하고 분리하여 GQD용액을 제조하였다. 제조된 GQD는 고순도의 그래핀으로 이루어져 있으며 그 크기와 두께가 일정한 나노 입자인 것으로 밝혀졌다. GQD-에탄올 용액은 아무 처리를 하지 않은 상태에서는 거의 광 발광하지 않았으나 초음파 처리를 추가적으로 실시한 후에는 발광 스펙트럼을 보였다. XPS 분석을 진행한 결과, 초음파 처리 공정을 GQD에 진행한 후 GQD 표면이 산화된 것으로 드러났다. 따라서 GQD 용액의 발광 원인은 표면 산화로 추측된다. 세포독성 실험 결과, GQD는 낮은 세포독성을 보였으며 GQD를 세포에 주입한 후에도 세포가 파괴되지 않고 정상상태를 유지함을 관측하였다. 따라서 본 연구를 통해 제조된 GQD는 향후 바이오 분야에 이용될 수 있을 것으로 기대된다.
