Dynamic trafficking of AMPA-type glutamate receptor (AMPA-R) in neuronal cells is a key cellular mechanism for learning and memory in the brain, which is regulated by AMPA-R interacting proteins. Although several postsynaptic proteins that interact with the AMPA-R have been identified as potential regulator of AMPA-R trafficking, the precise mechanism for the modulation of AMPA-R trafficking in and out of synapse is still under investigation. Here, we performed the precise functional study of LARGE, novel interactor of AMPA-R, revealed through our proteomic analysis. Also, we have identified Cav subunit that mediated the interaction between $Ca_v$ and AMPA-R, which can examine their functional association in postsynaptic membrane. Our functional study of LARGE, an intellectual disability-associated protein, indicates that the fine-tuning of AMPA-R trafficking by it as a molecule linking Hebbian and homeostatic synaptic plasticity, which is critical for memory stability is critical for memory formation in the brain. Next, The functional association of Cav2 with AMPA-type glutamate receptor (AMPA-R) in the postsynaptic membrane was mentioned in our previous study. Here, we have identified Cav subunit ($\alpha 1$) that mediates the interaction between Cav2 and AMPA-R through a series of biochemical experiments. which can increases the cell surface localization of AMPA-R.
Altogether, our study identified molecular mechanisms underlying the regulation ofA MPA-type glutamate receptor trafficking by its interacting proteins for synaptic plasticity and memory.
중추신경계의 후 시냅스에서 빠른 흥분성 신경전달을 중계하는 앰파-타입 글루타메이트 수용체의 신경 세포 표면과 시냅스로의 조절은 학습과 기억에 중요한 세포학적 기작이다. 이러한 조절은 이 수용체와 상호작용하는 다양한 단백질에 의해서 일어나게 된다. 현재 앰파-타입 글루타메이트 수용체와 상호작용하는 몇가지 단백질들이 밝혀졌지만 그것들이 어떻게 이 수용체를 세포막으로 이동시키고 시냅스로 표적 되게 하는지에 대한 정확한 기작은 밝혀져 있지 않다. 여기에서, 우리는 첫번째로 앰파-타입 글루타메이트 수용체와 상호작용하는 단백질로 밝혀진 라지 단백질에 의해 이 수용체가 신경세포 표면과 시냅스에서 조절되는 정밀한 기능적 기작을 연구하였다. 또한 두 번째로 시냅스에서 앰파-타입 글루타메이트 수용체와 칼슘채널간의 기능적인 연관성에 관한 연구를 바탕으로 이 두 단백질의 기능과 관련이 있는 단백질-단백질간의 상호작용에 대한 기작에 대해 연구하였다. 심각한 지적장애와 관련이 있는 라지 단백질에 대한 우리의 기능적 연구는 골지에서 라지 단백질에 의한 앰파-타입 글루타메이트 수용체의 미세한 조정이 뇌의 해마와 관련된 기억에 중요하다는 것을 밝혔다. 따라서 라지 단백질은 지적장애와 관련된 뇌질환의 병리생리학에 대한 새로운 통찰력을 제공하면서 두뇌의 기억과 그 안정성에 중요한 두 가지 시냅스 가소성인 헤비안과 항상성 시냅스 가소성을 연결하는 중요한 분자임을 확인하였다. 다음으로 우리의 이전 연구에서 후 시냅스에서 앰파-타입 글루타메이트 수용체와 칼슘채널의 기능적 연관성에 대해 보고 했다. 여기서 우리는 일련의 생화학 실험을 통해 이 둘간의 상호 작용에 관여하는 칼슘채널 단위체 (알파-1) 을 확인하였고 기능적인 분석에서 칼슘채널 알파-1 단위체가 수용체의 세포 표면의 위치를 증가 시키는 것을 확인하였다.
종합적으로 이야기해서 본 연구는 시냅스 가소성과 기억현상을 위한 앰파-타입 글루타메이트 수용체의 이동에 관여하는 단백질들의 역할을 분자적 수준에서 규명하였다.
