Since cancer spheroids can mimic the characteristics of in vivo tumor such as cell-cell, cell-ECM interactions and biochemical gradients, it is emerging as a next-generation platform for various applications including high-throughput drug screening. Currently, numerous methods for cancer spheroid culture have been developed. Conventional methods commonly utilize hydrophilic surface, while our research group has developed a hydrophobic polymer surface which can induce more tumorigenic cancer spheroids. On this point, here, we hypothesize that surface wettability has a significant effect on the characteristics of cancer spheroids. Hydrophilic polymer poly hydroxyethyl methacrylate (pHEMA) and hydrophobic polymer poly cyclohexyl methacrylate (pCHMA) were deposited via initiated Chemical Vapor Deposition (iCVD), and the cancer spheroids induced by these two surfaces were characterized from various perspectives. In particular, we explain tumorigenicity and the related mechanism in relation to the inner structure, thereby suggesting the ideal structure of cancer spheroids.
삼차원 배양을 통해 형성된 암세포 스페로이드는 세포와 세포 또는 세포와 세포외기질의 상호작용, 생화학적 구배 등 실제 체내의 암세포의 특성을 모사할 수 있기에, 고속 약물 스크리닝과 같은 다양한 연구에 활용할 수 있는 차세대 플랫폼으로 떠오르고 있다. 현재 삼차원 배양을 위한 많은 방법들이 개발되었으며, 기존의 표면 자극을 통해 암세포 스페로이드의 형성을 유도하는 방법들은 친수성 표면를 사용하고 있는 반면에, 본 연구 그룹에서는 소수성 고분자 표면을 통해 보다 더 악성의 암세포 스페로이드를 형성하는 방법을 개발한 바 있다. 이 점에 착안하여, 본 학위논문에서는 표면의 젖음성이 그 위에서 형성된 암세포 스페로이드의 특성에 중요한 영향을 끼친다고 가정하였다. 기상화학증착법 (iCVD)를 통해 친수성 고분자인 poly hydroxyethyl methacrylate (pHEMA)와 소수성 고분자인 poly cyclohexyl methacrylate (pCHMA)를 증착하였으며, 이 두 가지 표면 위에서 형성된 암세포 스페로이드를 다양한 관점에서 비교 및 분석하였다. 특히 암세포 스페로이드의 종양유발성 및 관련 메커니즘을 내부 구조와 연관 지어 설명하고자 하였으며, 이를 통해 이상적인 암세포 스페로이드의 구조를 제안할 수 있었다.
