A study of the redox properties of tamoxifen, one of the most common drug for treatment of breast cancer, with Density Functional Theory is presented here. Prediction of redox potential is exceedingly complicate and requires deeper understanding and careful consideration of factors which affect redox behavior of tamoxifen including the influence of ion paring and deprotonation. We carried out DFT calculations to gain oxidation potentials of each species. To consider ion pairing effect of tamoxifen derivatives in solvent, counterion, [$B(C_6F_5)_2F_2]^{-}$, is introduced. In the case of 2a and 2b, the computed reaction energy profiles are in fair agreement with experimental oxidation potentials. Otherwise we provide redox potentials of 2c which take into account the deprotonation reaction that occurs during the oxidation process because of the low pKa value of hydroxyl group of 2c structure. Meanwhile, oxidation property of tamoxifen can be improved by attaching cymantrene tag and electron donating group to the molecule, especially ethylene branch of the tamoxifen derivatives.
타목시펜이 엔독시펜으로 활성화 되는 반응을 산화 전위를 이용하여 분석하였다. 다양한 실험에서 메탈로신 태그는 엔독시펜으로의 산화 전위를 낮추는 역할을 한다는 것이 보고되었다. 산화 과정에서의 전이 금속 화합물과 라디칼 이동의 역할을 분석하기 위해 각 종에 대한 산화-환원 전위를 계산하였다. 우리는 디클로로메탄에서 시만트렌 태그된 타목시펜의 산화-환원 전위를 예측하기 위해 반대 이온 결합 구조가 중요한 역할을 한다는 것을 입증하였다. 한편, 전하 분포도 분석으로부터 높은 산화-환원 전위를 제어하는 가장 효과적인 자리가 에틸렌 가지라는 것을 관찰하였다. 또한, 탈양성자화 반응이 일어나는 경우 이를 고려하여 각 종들의 산화-환원 전위를 계산하였다. 우리는 이 연구를 통해 용액내에서의 산화-환원 쌍의 전위를 예측하는데에 반대 이온 및 탈양성자화 반응의 중요성을 입증하는 한편, 타목시펜의 산화 메커니즘에 대한 심도 있는 고찰을 통해 엔독시펜으로 쉽게 활성화 할 수 있는 전략을 제시하였다.
