This study proposes a mathematical model for the continuous fermentation process with ex-situ butanol recovery (ESBR) for biobutanol production using glucose and xylose, and performs model-based optimization and sensitivity analysis. The ESBR system is an integrated system of a fermenter and adsorption columns, alleviating the inhibitory effect on cell growth and production due to butanol toxicity through butanol recovery by adsorption. The system is difficult to operate and control intuitively owing to periodic switching of adsorption columns and complicated dynamic characteristics of the fermentation process. Thus, it is necessary to improve the operation stability and economic efficiency of the system through model-based optimization. Although the modeling and optimization of the bioethanol production process have been studied so far, modeling and optimization of the biobutanol production process have been rarely studied, and in particular, the modeling study on the process for producing biobutanol by simultaneous utilization of glucose and xylose has not yet been conducted. Therefore, this study proposes a concentration-dependent weighting factor to describe simultaneous utilization of glucose and xylose and develops a dynamic model of continuous biobutanol fermentation process with ESBR. A systematic approach has also been introduced for reliable model parameter identification. Based on the constructed model, multi-objective optimization is performed to improve the butanol productivity and to reduce the sugar loss. The ESBR system converges to cyclic steady state (CSS) due to periodical switching of adsorption columns, thus a simultaneous approach is used for optimization at the CSS. Finally, the roles and influences of each parameter are analyzed through sensitivity analysis under various optimal operating conditions to provide insights into optimal operation strategy by improving understanding of overall process dynamics.
본 연구에서는 포도당 및 목당을 이용한 바이오부탄올 발효/흡착 통합공정의 수학적 모델을 제안하고 모델 기반의 최적화 및 민감도 분석을 수행한다. 흡착 시스템이 통합된 바이오부탄올 연속 발효공정은 발효기와 외부 흡착탑이 결합된 형태로, 흡착에 의한 부탄올 회수를 통해 부탄올 독성에 의한 세포 성장 및 생산 저하를 완화한다. 이러한 통합공정은 주기적인 흡착탑 교체와 발효공정의 복잡한 동적 거동으로 인해 직관적으로 시스템을 운전하고 제어하기가 어렵고, 따라서 모델 기반의 최적화를 통한 시스템의 운전 안정성 및 경제성 향상이 필요하다. 현재까지 바이오에탄올 생산 공정에 대한 모델링 및 최적화 연구는 많이 진행되어 왔지만 바이오부탄올 생산 공정에 대한 모델링 및 최적화 연구는 거의 이뤄진 바가 없고, 특히 포도당 및 목당을 동시 섭취하여 바이오부탄올을 생산하는 공정에 대한 모델링 연구는 아직 수행되지 않았다. 따라서 본 연구에서는 포도당 및 목당의 동시 섭취를 모사하기 위한 농도 의존적 가중 인자를 제안하고 이를 이용하여 발효/흡착 통합공정의 동적 모델을 개발한다. 또한, 신뢰성 높은 모델 파라미터 식별을 위해 체계적인 접근법이 도입된다. 구축된 모델을 기반으로 부탄올 생산성을 향상시키고 당 손실을 저감하기 위해 다목적 최적화를 수행하여 최적의 운전 조건들을 찾는다. 발효/흡착 통합공정은 주기적인 흡착탑 교체로 인해 주기적 정상 상태로 수렴하는데, 이러한 주기적 정상 상태에서의 최적화를 위해 동시 접근법이 사용된다. 최종적으로 다양한 최적 운전 조건에서의 민감도 분석을 통해 각 파라미터의 역할 및 영향을 분석하여 전반적인 공정의 동적 거동에 대한 이해를 향상시킴으로써 최적 운전 전략에 대한 통찰을 제공하고자 한다.
