Plant hairy root cultures of Catharanthus roseus and Lithospermum erythrorhizon were performed to produce plant-derived secondary metabolites such as shikonin and indole alkaloids. Various hairy root cell lines were derived from two types of C. roseus plants (Catharanthus roseus cvs. Little Linda (LL) and Little Delicata (LD)) by transforming using Agrobacterium rhizogenes ATCC 15834. Hairy root clones derived from LD plants and LL plants showed contrasting morphologies. The hairy root derived from LD plants showed the characteristics of thick, straight, and fragile branches. In contrast to that, the hairy root derived from LL plants showed the characteristics of thin, coil-like, and unfragile branches. The production of alkaloids and root growth between hairy root clones were affected by their morphological characteristics. Hairy root clone LL 15 which showed the excellent growth and alkaloids productivity was selected for this study. The optimization of environmental condition was carried out both in C. roseus and L. erythrorhizon hairy root cultures. When sucrose was used as a carbon source in L. erythrorhizon hairy root culture, the maximum shikonin production and hairy root growth were obtained. However, when glucose or fructose was used instead, hairy root growth was severely inhibited. In contrast to this, hairy root culture of C. roseus, either glucose or fructose was a suitable carbon source as well as sucrose. Plant hairy root cultures of Lithospermum erythrorhizon were carried out to produce shikonin derivatives by employing in situ extraction with nhexadecane in a shake flask and a bubble column bioreactor. Over 95\% shikonin produced was recovered in the n-hexadecane layer. In flask cultures the concentration of shikonin with n-hexadecane extraction was 3 times than that obtained without extraction. To test the effect of elicitation on shikonin production and secretion, Penicillium species homogenate which was isolated from contaminated plant cell culture, was adopted in this study as fungal elicitor: Shikonin production reached its maximum when the concentration of the fungal elicitor was around 0.02 g/l. However, in the culture of hairy roots, the increase of shikonin induction by fungal elicitation was not significant in comparison with that of normal cell culture in the earlier report. In the two phase bubble column reactor, 572.6 mg/l of shikonin and 15.6 g/l of dry cell mass were obtained after 54 days of culture. Shikonin was produced at a constant level of 10.6 mg/L day during this period. In order to enhance the alkaloid production and secretion, the influences of the addition of XAD-7, DMSO, and fungal elicitor as the physical and biochemical stress, and their synergistic action on the production of catharanthine and ajmalicine were investigated. Among various Amberlite resins (XAD-2, XAD-4, and XAD-7), the maximum production of catharanthine and ajamlicine was obtained when XAD-7 was added to halfstrength SH liquid medium. DMSO treatment with in situ product adsorption by XAD-7 for the release of intracellularly stored indole alkaloids appears to be appropriate for the secretion of alkaloid from the cells. In addition, in situ adsorption and fungal elicitation had a positive effect on alkaloid production and secretion. By combining product adsorption sequentially with above useful techniques, the chances of favorable effects improve substantially. Total production of catharanthine was 67 mg/l, which was about five times higher than that of control culture. Total production of ajmalicine was enhanced up to 30.15 mg/l, which was 3.7 times higher than that of control culture. Fed batch culture of L. erythrorhizon hairy root was carried out by controlling sucrose concentration and medium conductivity in shake flask and a modified stirred tank reactor. In addition, for the effective product recovery from the culture, in situ adsorption by XAD-2 was also applied. When sucrose was used as a carbon source, the highest shikonin production and hairy root growth were obtained. Alternating feeding of sucrose enhanced the productivity of shikonin derivatives remarkably. As the resin amount increased up to 1.5 g/l, shikonin production was enhanced by efficiently removing shikonin produced and other products which might be inhibitory to cell growth. Most of shikonin produced was recovered in XAD-2 resin (Over 99\%). Using hairy root culture in a modified stirred tank reactor, the shikonin productivity and hairy root growth rate on the average increased to 9.34 mg/l.day and 0.49 g dry weight/l.day, respectively.
본 실험은 식물의 조직 배양을 통해 얻어지는 유용한 이차대사물을 효과적으로 식물체로 부터 생산 및 동시 분리하기 위한 효과적인 분리 방법의 개발과 또한 이와같은 분리가 식물체의 성장과 대사에 미치는 현상을 알아보고자 하였다. 그 모델 식물체로서 여러가지 유용한 항암제를 함유한 일일초 (Catharanthus roseus)와 화장품과 의약품의 원료로 이용되는 시코닌을 함유한 자초 (Lithospermum erythrorhizon)의 모상근을 이용하였다. 이차대사물에 대한 생산성이 높은 것으로 알려진 모상근 유도 실험을 수행하였다. 두 종류의 일일초 (Catharanthus roseus cvs. Little Linda (LL) and Little Delicata (LD))로부터 토양세균 Agrobacterium rhizogenes ATCC 15834를 이용하여 여러 모상근이 유도되었다. LL 식물체와 LD 식물체로부터 유도된 모상근은 형태적으로 서로 다른 특징을 갖고 있었으며 알칼로이드의 생산성과 모상근의 성장도 서로 다른 특징을 보였다. 모상근으로부터의 이차대사물의 분리및 생산 실험의 수행을 위해 모상근 LL 15가 모델로 선정되었으며 알칼로이드 생산성이 다른 유도된 모상근에 비해 뛰어난 것으로 나타났다. 유도된 일일초 모상근 LL 15와 자초 모상근에 대한 배양조건을 살펴보았다. 특히, 단당류의 식물세포배양에서의 이용은 탄소원 제어를 통한 유가식 배양에는 필수적이다. 자초 모상근의 탄소원 실험에서는 sucrose를 이용하였을 때 가장 좋은 생산성을 보였으나 단당류에 대한 모상근 성장의 저해가 심각하였다. 그러나 일일초의 경우는 fructose및 glucose 에 대한 모상근 성장의 저해가 없었다. 자초와는 달리 일일초 모상근에 있어서 glucose와 fructose는 sucrose와 함께 적당한 탄소원임이 밝혀졌다. n-Hexadecane을 이용한 동시추출 실험이 플라스크 배양과 기포탑 생물반응기에서 수행되었다. 95\% 이상의 시코닌이 n-hexadecane 층으로 회수 되었다. 플라스크 배양실험에서 동시추출을 이용한 시코닌 생산이 동시추출을 하지 않은 배양에 비해 약 3배 가량 증가함을 알수 있다. 식물세포 배양에서 생산성의 향상을 위해 자주 이용되는 Elicitor를 그 첨가양을 변화시켜 자초 모상근에 적용시킨 결과 시코닌 생산이 0.02 g/l의 fungal elicitor를 첨가하였을 때 가장 높았다. 그러나 자초 모상근에 있어서의 Fungal Elicitation에 의한 시코닌의 유도는 탈분화세포 배양에 비해 Elicitation 효과가 작았다. 이는 분화된 u}캣걘셈? 유전적 안정성에 기인한다고 생각된다. 동시추출을 이용한 플라스크 배양의 결과를 토대로 이를 이상 기포탑 반응기 (Two-phase bubble column reactor) 라는 새로운 개념의 시스템에 적용한 결과 54일 동안 572.6mg/l의시코닌이 생산되었으며, 15.6 g/l의 건조중량이 얻어졌다. 또한 이 기간동안의 시코닌 생산성은 하루평균 10.6 mg/L 정도로 생산되었다. 알칼로이드 생산실험에서 알칼로이드의 생산성과 분리효율을 높이기 위해 물리적 생화학적 자극으로써 흡착제인 XAD-7, Permeabilizing agent로서 Dimethyl sulfoxide (DMSO), 그리고 Fungal Elicitor에 대한 카타란틴 (Catharanthine)과 아즈말리신(Ajmalicine) 생산효과를 살펴보았다. 카타란틴은 항암제인 vinblastine과 vincristine의 전구체이며 아즈말리신은 혈압강하제로 이용되는 의약품이다. 여러 Amberlite 수지를 첨가해 본 결과 XAD-7이 알칼로이드 생산에 가장 좋은 효과를 나타내었다. 알칼로이드의 분리효율을 높이기 위해 동시흡착과 함께 DMSO를 첨가해 본 결과, 모상근으로부터 상당량의 알칼로이드에 대한 분리효과가 있었다. 더불어 동시추출과 Fungal Elicitation도 알칼로이드 생산에 긍정적 효과를 나타내었다. 마지막으로 이 세가지 유용한 기술을 순차적으로 모상근 배양에 적용시켜 보았다. 그 결과 카타란틴의 분리는 control 배양에 비해 3.5배 이상 증가하였다. Sucrose 농도와 배지의 전도도(conductivity)를 제어하여 자초 모상근의 유가식 배양을 플라스크 배양과 개량된 교반반응기에서 각각 수행하였다. 더불어 효과적인 시코닌의 분리를 위해 XAD-2를 이용한 동시흡착도 함께 수행하였다. Sucrose가 탄소원으로 사용되었을 때, 모상근의 성장이 가장 좋았다. Sucrose의 공급 방식의 변화는 시코닌 생산을 향상시키는 효과적인 방법으로 이용되었다. 흡착 실험에서 1.5 g/l의 XAD-2 수지를 첨가한 결과 상당한 시코닌 생산의 증가를 보았다. 99\% 이상의 시코닌이 흡착제에 흡착되었다. 산소제어의 용이함을 위해 고안된 고정화용 교반 반응기에서의 모상근 배양을 통해 얻어진 평균 시코닌 생산과 모상근 생산 속도는 각각 9.34 mg/l.day 및 9 g dry cell/l/day 였다.
