A microorganim was isolated from soil for simultaneous biodegradation of BTX and identified as Pseudomonas fluorescence BE103. Using Pseudomonas fluorescence BE103, biodegradation of aromatic hydrocarbons by cometabolism was characterized and their metabolic pathway was analyzed. Pseudomonas fluorescence BE103 utilized benzene and toluene as growth substrates, but p-xylene by cometabolism. Cometabolism of p-xylene was dependent on the presence of growth substrate and active biomass. The metabolic product from cometabolism of p-xylene was identified as dimethylcatechol by LC/MS. From the measurement of enzyme activity for ring activation and ring fission that determine the metabolic pathway of aromatic hydrocarbons, the metabolic pathway for BTX degradation in Pseudomonas fluorescence BE103 was analyzed. The pathway was similar to TOD pathway. Cometabolism of p-xylene might be attributed to the broad substrate specificity of ring activation dioxygenase and the absolute substrate specificity of ring fission dioxygenase. Simultaneous biodegradation of BTX was not accomplished by mixed cultivation with Pseudomonas putida mt-2. For simultaneous biodegradation of BTX, the development of the microbe is required.
BTX 의 동시 생분해를 위해서 토양으로부터 분리한 Pseudomonas fluorescence BE103 을 이용하여, cometabolism 에 의한 방향족 탄화수소의 생분해와 그 대사 경로를 분석하였다. Pseudomonas fluorescence BE103 은, benzene 과 toluene 은 탄소 및 에너지원으로 이용하나 p-xylene 은 cometabolism 을 통하여 이용하여 대사 산물을 축적시켰다. p-Xylene 의 cometabolism 은 benzene, toluene 과 같은 탄소 및 에너지원의 존재와 활성을 가지는 균체량에 의존적으로 일어났다. 또한, p-xylene 의 이용으로부터 축적되는 대사 산물은 LC/MS 분석을 통하여 dimethylcatechol 임이 확인되었다. 방향족 탄화수소의 대사 경로를 결정짓는 ring activation 과 ring fission 단계의 효소활성을 측정하여, Pseudomonas fluorescence BE103 의 BTX 대사는 tod pathway 와 유사한 경로로 이루어짐을 알 수 있었다. p-Xylene 의 cometabolism 은 ring activation dioxygenase 의 광범위한 기질 특이성과 ring fission dioxygenase 의 높은 기질 특이성에 의한 현상으로 생각된다. BTX 의 동시 생분해를 위해 toluene 과 p-xylene 을 탄소 및 에너지원으로 이용하는 Pseudomonas putida mt-2 와 혼합 배양을 수행하였으나, 이 때도 대사 산물이 축적되었다. 따라서 BTX 의 동시 생분해를 위해서는 새로운 균주의 개발이 요구된다.
