A bacterial strain capable of degrading pyridine was isolated from a retort water treatment column(RWTC) by enrichment culture techniques. The micro organism was aerobic, Gram-positive, pleomorphic, nonsporeforming and had a subpolar flagellum. The strain contains LL-diaminopimelic acid, galactose, and glycolyl residues in the cell wall. The G+C content of DNA was 72.5 mole %. The major manaquinone was MK-8($H_4$) and the cellular fatty acid composition was complex type. Based on the morphological, physiological and chemotaxonomic characteristics, the strain was identified as Pimelobacter sp. The microorganism could oxidize pyridine for energy, nitrogen and carbon sources. The nitrogen in pyridine was released as ammonium. The microorganism could decrease TOC(total organic carbon) of the retort waters and catabolize other aromatics. These results indicates this organism could be well adapted to retort waters. When pyridine was present, catalytic activity to pyridine was induced. Pyridine has the effect of changing the fatty acid composition of the membrane. The result of these changes was an increase in the degree of saturation of lipids probably in order to compensate an increase of fluidity of the membrane induced by the pyridine. In small inoculum size, readily available nutrients stimulated growth rate but the specific degradation rates were inverse to the growth rate. Ammonium ion increased both growth rate and specific degradation rate. In large inoculum size, the specific degradatrion rate was not decreased by these nutrients, and ammonium ion had no effect on pyridine degradation. In immobilized cells degradation rate was limited by diffusion at low concentration of pyridine, but at higher concentration of pyridine immobilized cells were more tolerant than free cells. On semicontinuous cultures immobilized cells could maintain catalytic activity for 70 hours during six times of the replenishment of the media at 1 g/L of pyridine conc. and retained catalytic activity for 180 hours during three times of the replenishment at 5 g/L of pyridine concentration.
RWTC 로 부터 농화 배양(enrichnment culture)법을 이용하여 pyridine 자화 미생물을 분리하였다. 그램 양성균중 활성이 뛰어난 한 균을 선별하여 실험을 계속하였다. 이 균주는 호기성이었으며, 형태변화를 일으키는 미생물이었다. 화학 동정 결과 LL-DAP, 갈락토스, 글리콜릴기, MK-8($H_4$), 복잡한 지방산 형태를 가지고 있었으며, DNA 의 G+C content 는 72.5 % 였다. 이결과와 생리적 특징를 이용하여 이균주는 Pimelobacter sp 로 동정되었다 촉매능은 pyridine에 의해 유도되어 에너지원, 탄소원 및 질소원으로 이용하여 성장할수 있 었으며 남은 질소는 암모니아형태로 방출하였다. 다른 방향족 화합물들에 대한 자화능이 뛰어 났으며 retort water 의 총유기탄소(Total Oganic Carbon)를 감소시킬수있음을 알수있었다. 4 g/L 이상에서는 독성으로 인해 성장을 하지 않았으며, 농도가 높아짐에 따라 성장 효율이 낮아졌다. pyridine에 의한 독성을 방지하기위해 세포막의 지방산조성을 변화시켰다. 포도당에서 자란 세포에 비해 pyridine에서 자란 세포가 막 유동성이 낮았으나, pyridine 농도변화에 의한 막 유동성의 변화는 기존에 발표된 결과와는 다른 양상이었다. 접종양이 적을때는 효모 추출물등의 영양소에 의해 pyridine의분해가 저해를 받았으며 비성장속도와 비소모속도는 서로 반대되는 경향이 있었다. 암모니아는 초기에 성장속도와 소모속도를 동시에 증가시키는 경향이 있었다. 하지만 접종량이 많을 때는 이들에 의한 억제는 보이지 않았으며, 고정화 세포에서는 억제 효과가 감소됨을 알수있었다. 세포농도의 증가에 의해 세포의 비성장속도는 거의 변하지않았다. 고정화 세포는 3 g/L 까지의 농도에서는 기질 확산속도에 의해 pyridine 분해가 그렇지않은 세포에 비해낮았으나, 농도가 증가할때는 고정화 시키지 않은 세포에 비해 상대적으로 적은 저해를 받는겄을 알수있었다. 반연속 배양결과 1 g/L pyridine 농도에서 여섯번의 반복으로도 분해능에 변화가 없었으며, 5 g/L 농도에서는 180 시간 동안 촉매능이 유지됨을 알수있었다.
