The suppressive effect of epidermal growth factor(EGF) on drug-metabolizing activity was investigated in primary culture of mouse hepatocytes. EGF decreased P-450IA-specific ethoxyresorufin O-deethylase(EROD) activity, P-450IIB-specific penthoxyresorufin O-dealkylase(PROD) activity, and P-450IIIA-specific erythromycin N-demethylase(ERDM) activity significantly. EGF(0.1-10 μg/ml) reduced EROD activity in dose and time-dependentmanner during 3 day cultures. At the lowest concentration, 0.1 μg/ml, EGF could decrease EROD activity meaningfully. The suppressive effect of EGF appeared at 4 hr, and reached maximum within 24 hr. PROD activity was also inhibited by EGF in 3 day cultures, but at relatively high concentration of EGF (10μg/ml). In contrast to EROD, PROD activity showed weak dose-dependency of EGF treatment. Dephosphorylation experiments showed that the inhibition of EROD and PROD activity was not resulted from phosphorylation of microsomal proteins. ERDM activity was also subject to EGF. In the case of PROD and ERDM, EGF reduced the basal activity of enzymes as well as chemical(2mM phenobarbital and 10 μM dexamethasone, respectively)-induced activity, while the basal EROD activity was elevated slightly by EGF in 3 day cultures. Western immunoblot analysis with P-450IA- and P-450IIB-specific monoclonal antibodies showed that the inhibition of phenotypic enzyme activities was found to be due to the decrease of amount of proteins. This change of protein amount was resulted from reduction of mRNA expression by EGF, which was proved in RNA slot blot analysis.
본 연구에서는 간세포 재생 과정에서 중요하다고 알려진 Epidermal Growth Factor(EGF) 가 간세포 내의 약물 대사능에 어떤 영향을 미치는 가를 mouse 의 일차배양 간세포를 이용하여 조사하였다. P-450 발현 조절 기작의 다양성을 고려하여, P-450IA에 특이적인 ethoxyresorufin O-deethylase(EROD), P-450IIB에 특이적인 penthoxyresorufin O-dealkylase(PROD), 그리고 P-450IIIA에 특이적인 erythromycin N-demethylase(ERDM) 의 세 종류의 효소를 선택, 그 활성도가 EGF에 의해 어떻게 변화하는지의 양상을 보았다. 그 중 EROD의 활성도는 EGF에 의해 뚜렷한 선별성을 나타내며 감소하였고 나머지 두 효소에서도 유의성 있는 감소가 나타났다. 이러한 P-450 발현 감소의 기작을 좀 더 자세히 조사하기 위해 단백질 수준의 Western blot analysis와 mRNA전사 수준의 RNA blot analysis 를 수행하였다. 그 결과 EGF는 P-450IA1 의 단백질 양과 mRNA 양을 현저히 감소시키고 P-450IIB1 의 경우에도 효소 활성도의 억제 결과와 일치하는 정도의 단백질 양의 감소와 mRNA 감소가 관찰되었다. 한편, P-450 감소가 단백질의 인산화에 의해서 일어날 수도 있다는 가능성을 조사하기 위해 탈인산화 실험도 수행해 보았으나 EGF 에 의한 약물 대사능 감소는 탈인산화에 의해 영향을 받지 않는 것으로 나타났다. 이 결과로부터, EGF 는 P-450 발현을 억제하며, 그 기작은 전사 수준에서 일어난다는 결론을 얻을 수 있다.
간세포 재생 과정중에 약물 대사능의 억제 현상은 오래 전부터 알려져 있었으나 그 기작에 대한 보고는 거의 없었다. EGF 가 간 재생 과정에서 중요 인자라는 가설이 사실이라면 이 실험은 간 재생 과정의 약물 대사능 억제가 EGF 수용체를 통해서 일어날 수 있다는 간접적인 증거가 될 것이다.
