The pho5 gene of S. cerevisiae encodes the repressible acid phosphatase(rApase) which is regulated by the concentration of Pi in a medium. Introducting the pho5 gene into tobacco plants and allowing sebsequent expression under the control of CaMV 35S promotor, it is expected to make transgenic plants resistant to Pi deprivation in acidified soil. For this purpose, plasmid pBSW1 was constructed by the insertion of 1.5 kb DNA fragment containing the entire open reading frame of pho5 gene into T-DNA region of pBKS1-1, a binary vector. Tobacco cells were transformed by co-incubating with A. tumefaciens cells carryng pBSW1. Transformed cells were regenerated into natural plants which have the T-DNA region of pBSW1 in their genome.
효모의 pho5 유전자는 외부 인산 농도에 의해 조절되는 repressible acid phosphatase를 암호화하고 있다. 담배조직내로 pho5 유전자를 도입하여 CaMV 35 promotor의 조절하에 발현시키므로써, 산성화된 토양에서 인산결핍에 대한 저항성을 지닌 "transgenic plant"를 유도할수 있을 것으로 기대된다. 이러한 목적으로, pho5 유전자의 전체 open reading frame을 포함하는 1.5 kb DNA 절편을 pBKS1-1의 T-DNA 영역내에 삽입시켜 플라스미드 pBSW1을 만들었다. 그리고, pBSW1을 옮기는 A. tumefaciens와 담배의 잎조각을 함께 배양하여 담배세포를 형질전환 시켰다. 형질전환된 세포는 정상적인 식물체로 재분화되었으며 그 식물체의 게놈내에 pBSW1의 T-DNA를 포함하고 있음이 확인되었다.
