A method for preparing a respectable quality streptokinase in a convenient manner was described. Streptokinase is important for enzyme theraphy, because it lyses blood clots by activating plasminogen to the fibrinolytic enzyme, plasmin. For the crystallography to determine its tertiary structure, the preparation of pure streptokinase is needed. Human plasminogen was purified from Cohn fraction III by the affinity chromatography with L-lysine-substituted Sepharose CL-4B. This plasminogen was used as a ligand for streptokinase affinity chromatography. PBS (pH7.4) and Glycine (pH2.5) were used as washing and elution buffer, respectively. To obtain a single peak, ε-amino-N-caproic acid (ε-ACA) was added to both washing and elution buffers. The function of ε-ACA is the prevention of streptokinase fragmentation during the plasminogen activation.
To study the role of Gly 24 in streptokinase, mutant streptokinases, His 24, Ala 24 and Glu 24, were purified and tested for specific activity and protease activity with various artificial substrates. The specific activity of Ala-SK was about 76.6% of Gly-SK, and His-SK and Glu-SK lost nearly all activator activity. And any protease activity against substrates tested so far has not been detected in wild-type and mutant streptokinases.
효소 제제로써 중요한 스트렙토키나아제의 3차 구조 결정을 위해 필요한 한 과정으로 높은 순도를 가진 단백질 분리 방법이 개발되었다. 사람의 플라즈미노겐은 Cohn Fraction III로부터 L-lysine-substituted Sepharose CL-4B를 이용한 친화성 크로마토그래피로 분리하였고, 이 플라즈미노겐은 스트렙토키나아제의 친화성 크로마토그래피의 리간드로써 사용되어졌다. PBS (pH7.4) 와 Glycine (pH2.5) 이 각각 washing buffer과 elution buffer 로 사용되었다. 스트렙토키나아제의 순수분리를 위해서는 이 용액들 속에 ε-amino-N-caproic acid (ε-ACA)를 첨가해 주어야 한다. ε-ACA는 플라즈미노겐 활성화 시기에 발생하는 스트렙토키나아제의 절편화를 방지해 주는 역할을 한다.
스트렙토키나아제의 24번째 잔기인 Glycine의 역할을 연구하기 위해, Gly을 각각 His, Ala 그리고 Glu로 변화시킨 돌연변이 단백질들을 순수분리하여 플라즈미노겐 활성화 능력과 단백질 분해능력을 살펴보았다. Ala-SK는 플라즈미노겐 활성화 능력 (specific activity) 이 Gly-SK에 비해 약 76.6%정도로 나타났으며, His-SK와 Glu-SK는 그 능력이 거의 소실된 것으로 나타났다. 그리고, 위 단백질들의 단백질 분해능력에 대한 실험을 여러가지 인위적인 기질을 이용하여 살펴 보았으나 발견되지 않았다.
