The proteolysis of endo-β-1,4-glucanse (endoglucanase) in Bacillus megaterium and the study of cellulose binding domain were performed in this study. An endoglucanase gene of Bacillus subtilis was cloned previously from B. subtilis BSE616 in E. coli in our laboratory. Also, we have transferred this gene in B. megaterium ATCC 14945 which was cellulolytic negative strain. This intact endoglucanase (52kD) secreted into the culture medium was gradually cleaved to truncated form (33 kD) by extracellular protease. The intact endoglucanase, truncated endoglucanase and extracellular protease were purified to certify the proteolysis and domain structure. The extracellular protease was purified by Q-Sepharose, Sephadex and Hydroxyapatite chromatography. The molecular weight of the purified extracellular protease was 38 kD. The protease showed maximal activity at 55℃ and at pH 7.5. The protease that requires $Ca^{++}$ for its activity was metal protease. The intact endoglucanase was purified by affinity towards insoluble cellulose and Mono-Q Chromatography. The truncated endoglucanase was purified by gel filtration and chromatofocusing. The cleavage of the intact endoglucanase was investigated by using immunoblotting and reaction with purified extracellular protease. The secreted form of endoglucanase was 52 kD at early log phase, and this enzyme was further turther into the smallest form (33kd) by the extracellular protease. Silmilar results were obtained with chymotrypsin and trypsin. From the N-terminal sequencing data, it was concluded that direction of cleavage is from C-terminus. The molecular activities of these enzymes towards the soluble substrates were identical. However, the insolube substrate, Avicel, was hydrolyzed slowly by the intact endgoglucanase on the other hand the truncated endolucanase did not hydrolyze the Avicel. Isoelectric points (pI) of the intact and truncated endoglucanase were 6.2 and 5.6, respectively. Optimum pH and temperature were 5.5 and 60℃, respectively. Only the intact endoglucanase was adsorbed to Avicel. The practical maximum adsorption was about 20 mg/g of cellulose. At pH 6, maximum adsorption occurred. The adsorption was the highest below 5℃ and decreased with increasing temperature. For immobilization of β-glucosidase using cellulose-binding domain of endoglucanase, recombinant plasmid (pβCBD) for fusion protein was constructed. The fusion protein (80 kD) was well immobilized to Avicel and hydrolysis of substrate (cellobiose) was complete. β-Glucosidase-CBD fusion protein bound to Avicel retained full activity during 24 hours continuous operation at 4℃
섬유소를 이용한 에너지원의 개발을 위하여 섬유소 분해에 관여하는 효소들의 유전자및 효소의 특징에 대한 많은 연구가 진행되어 왔다. 이러한 연구의 일환으로서 섬유소 분해 효소의 하나인 endo-β-1,4-glucanase 의 정제및 Bacillus megaterium 의 세포의 단백질 분해 효소에 의한 proteolysis 와 아미노산 서열로 추정되는 domain 구조에 대하여 조사하였다.
B. megaterium 의 세포의 단백질 분해 효소를 3 단계의 chromatography 로 순수 분리하였고, 이 효소는 단일 분자로소 분자량은 38 kD 였으며, pH 7.5 와 55 ℃에서 가장 높은 활성을 보였다. 이 효소는 $Ca^{++}$ 이온을 요구하는 metal 계 단백질 분해 효소임을 알았다. Intact endoglucanase 는 불용성 기질 (Avicel) 에 대한 친화력을 이용한 2 단계의 chromatography 로, truncated endoglucanase 는 chromatoocusing 을 포함한 2 단계의 chromatography 로서 순수 분리하엿다. 각각의 분자량은 52 kD과 33 kD 였으며, pI 값은 각기 6.2 와 5.6 임을 확인하였다. Intact endoglucanase 의 절단은 세포의 단백질 분해 효소에 의하여 진행됨을 immunoblotting 과 직접 반응을 통해서 알아내었다. 생장 초기에는 52 kD 의 형태로 세포외로 유출되지만 시간이 감에 따라 세포의 단백질 분해 효소에 의해 33 kD 형태로 진행됨을 보았다. 이러한 현상을 chymotrypsin 과 trypsin 과의 반응에서도 일어나 각기 35 kD, 37 kD 의 형태로 절단되었다. 아미노산 서열 분석을 통해서 절단은 C-terminal 쪽에서 이루어짐을 확인하였다. 33 kD 부분이 효소 활성을 나타내는 곳이고 절단된 19 kD 부분에는 cellulose-binding domain 과 연결 부위가 포함되어 있음을 기존의 알려진 섬유소 분해 효소와 아미노산 서열을 비교하여 알게되었다. Intact endoglucanase 와 truncated endoglucanase 는 가용성 기질에 대한 활성이 같고, 최적 pH 는 5.5, 최적 온도는 60℃ 로 같고 pH 및 온도에 의한 활성 변화도 같은 양상을 나타내었다. 그러나 불용성 기질 (Avicel) 에 대해서는 intact endoglucanase 만이 활성을 보였고, 이는 절단된 19 kD의 부분이 기질 흡착에 관여한다는 증거가 되었다. Intact endoglucanase 는 기질 lg 당 약 20 mg 정도가 흡착되고 pH 6.5 ℃ 이하에서 가장 흡착능이 좋음을 확인하였다.
Cellulose-binding domain 을 이용하여 효소의 고정화의 가능성을 조사하기 위하여 β-glucosidase 유전자를 결합시켜 pβCBD 라는 유전자 운반체를 만들었고, 여기에서 전사된 80 kD 의 융합 단백질을 불용성 기질에 고정화시켰다. 대장균에서 발현된 융합 단백질은 손쉽게 고정화되었고, β-glucosidase 의 기질인 cellobiose 를 column 을 통해 반응시켜본 결과 glucose 로 95% 이상이 분해되었으며 thin-layer chromatography 로 확인하였다. 고정화된 융합 단백질은 4 ℃ 이하에서 24 시간동안 활성의 저하없이 동일한 활성을 유지함을 알아냈다. Cellulose-binding domain 을 이용한 효소의 고정화는 불용성 기질의 값이 싸고 고정화 과정이 간단하고 빠르며 4 ℃ 이하에서는 고정화된 효소의 유출이 없는 장점이 있다. 이 domain 의 연결부위는 3 차원적인 구조로 볼때 돌출되어있어서 유연성이 있고 효소의 활성을 떨어뜨리지 않으므로, 그 이용가치가 높다고 사료된다. 또한 섬유소 분해를 이용한 에너지원 개발의 측면에 있어서는 단백질 분해 효소에 의한 cellulose-binding domain 의 절단을 막는 것이 필수적이며, 따라서 glycosylation 이 가능한 효모나 단백질 분해 효소가 없는 균주를 숙주로 사용함이 바람직하다고 사료된다.
