As science and technology are developed in modern times, people are becoming exposed to many diseases. Because of this, research and development on the biosensor are getting a great attention. Among many researches on the biosensor, recent researches have focused on the application of Point-of-Care (POC) system, which is defined as simple medical diagnosis that can be conducted at the bedside.
So as to exploit the biosensor for POC system, several characteristics such as real time measure-ment, portability, and ease-of-use are required. These features can be achieved by utilizing surface plasmon resonance (SPR) technology. There are many different types of biosensor using SPR technology. In this thesis, a study on the waveguide type SPR sensor is included because this type has many advantages including min-iaturization and cost effective fabrication process.
Previously developed waveguide type SPR sensors are only based on the spectral modulation with keeping the incident angle of the light constant. That is because it is difficult to change the incident angle in a waveguide structure. These sensors have critical limitations in terms of the sensitivity because the incident angle is important parameter in surface plasmon resonance condition. However, this limitation can be easily overcome by using varifocal electrowetting effect of electrowetting (EW) lens. Therefore, in this research, a novel SPR waveguide sensor using both the spectral modulation and the angular modulation is proposed.
Firstly, numerical simulations are conducted for optimization of structure and analysis of angular properties. Subsequently, the SPR waveguide sensor is fabricated in accordance with optimization results and evaluated by measuring and calculating the normalized intensity, the operating range, and the sensitivity. The proposed SPR waveguide sensor shows that the minimum value of the normalized intensity is 0.1 ? 0.3, the operating range is 511.4 ? 576.2nm, and the sensitivity is 2160nm/RIU.
Compared to the sensitivity of the conventional sensing system based only on the spectral modula-tion, the sensitivity of the proposed sensing system is enhanced by up to 169.7%. This result demonstrates that the performance of the sensor can be further improved when using both the spectral modulation and the angular modulation.
현대에 들어 과학 기술이 점차 발전함에 따라, 사람들은 더욱 많은 질병에 노출되고 있다. 이 때문에, 바이오센서에 대한 연구 개발이 근래에 더욱 큰 관심을 받고 있다. 이러한 바이오센서에 관한 많은 연구들 중에서도, 최근 연구들은 침대와 같은 환자가 처치 받는 위치와 근접한 곳에서 간단하게 실시 가능한 현장 현시 검사 시스템(Point-of-Care: POC)에 초점을 맞추고 있다.
현장 현시 검사용(POC)으로 바이오센서를 이용하기 위해서는 실시간 측정, 휴대성, 사용 용이성 등의 특징이 요구된다. 이러한 특징들은 표면 플라즈몬 공명(Surface Plasmon Resonance: SPR) 기술을 이용하면 얻을 수 있다. 이러한 장점들을 바탕으로 한 SPR 기술을 이용한 바이오센서에는 많은 종류가 있다. 본 논문에서는 도파관 형태의 SPR 센서에 관한 연구가 포함되는데, 이는 도파관 형태의 센서가 다른 형태의 센서들에 비해 소형화 및 제작 가격 경쟁력 등에 있어서 큰 장점을 지니고 있기 때문이다.
이전까지 개발되었던 도파관 형태의 SPR 센서들은 스펙트럼 변조만을 기반으로 하여 동작하는데, 이는 빛의 입사각은 일정하게 유지하면서 넓은 범위의 파장 대역을 갖는 광원을 사용하는 것을 의미한다. 스펙트럼 변조만을 수행하는 이유는 도파관 구조 안에서 빛의 입사각을 변조시키기 어렵기 때문이다. 표면 플라즈몬 공명 조건에서 빛의 입사각은 중요 변수이기 때문에, 이와 같이 스펙트럼 변조만을 기반으로 한 센서들은 감도와 같은 부분에서 치명적 한계를 갖는다. 그러나 이러한 한계는 전기습윤 렌즈(EW-lens)의 전기습윤 초점 가변을 이용하면 쉽게 해결될 수 있다. 따라서, 본 논문에서는 스펙트럼과 입사각 변조 모두를 기반으로 한 새로운 SPR 도파관 센서가 제안된다.
먼저, 본 연구에서는 구조 최적화 및 각도 특성 분석을 위해 수치 시뮬레이션이 수행된다. 그리고 나서, 최적화 결과에 따라 SPR 도파관 센서가 제작되고, 정규화 강도, 동작 범위, 그리고 감도 등을 측정하여 성능 평가를 수행한다. 제안된 SPR 도파관 센서는 0.1 - 0.3의 정규화 강도, 511.4 - 576.4nm의 동작 범위, 그리고 2160nm/RIU의 감도를 보인다.
스펙트럼 변조만을 기반으로 하는 기존 시스템의 감도와 비교하여, 제안된 시스템의 감도는 최대 169.7%까지 향상된다. 이러한 결과는 스펙트럼과 입사각 변조 모두를 사용할 때 센서의 성능이 더 향상될 수 있음을 보여준다.
