Photothermal therapy is a minimally-invasive cancer treatment that uses nanoscopic agents together with near-infrared light (NIR) to destroy cancer cells. Although gold nanorods are the most commonly used photothermal agents, there are certain disadvantages that can decrease the effectiveness of photothermal therapy. Gold nanoparticles address the issues of gold nanorods, but lacks absorbance in the NIR region. Melanin is a well-known biopolymer with a wide absorbance, which extends into the NIR region. Herein, gold and melanin (Gold-melanin) hybrid nanoparticles are developed for photothermal treatment of lung carcinoma. The cancer-targeting functionality is enabled by conjugating epidermal growth factors (EGF) onto the surface of Gold-melanin nanoparticles by using a derivative of polyethylene glycol (PEG) as a spacer. The resulting nanoparticles, termed Gold-melanin@PEG-EGF, exhibit high biocompatibility, excellent dispersion stability in serum, and successful cellular internalization. By using near-infrared irradiation (808 nm, 500 mW $cm^{-2}$), Gold-melanin@PEG-EGF nanoparticles are able to selectively target and kill lung cancer cells after 30 min of irradiation in vitro, while leaving other cell lines (MCF-7 and HaCaT) unharmed. In addition, in vivo photothermal treatments show that Gold-melanin@PEG-EGF nanoparticles are able to suppress tumor growth after 5 min of laser irradiation (808 nm, 1.5 W $cm^{-2}$). The results show that Gold-melanin@PEG-EGF nanoparticles specifically bind to EGF-expressing cells and are able to generate localized hyperthermia to destroy these cells. These unique nanoparticles show great potential as a biocompatible photothermal agent that can effectively kill lung cancer cells
광열 요법은 암세포를 파괴하기 위해 근적외선과 함께 나노 약물을 사용하는 최소 침습적인 암 치료법이다. 금 나노 로드가 가장 일반적으로 사용되는 광열 제이지만, 광열 요법의 효과를 감소시킬 수 있는 단점이 있다. 또한 금 나노 입자는 금 나노 로드의 문제를 다루지만 근적외선 영역에서는 흡광도가 부족한 것으로 나타낸다. 반면에 멜라닌은 근적외선 영역으로 확장되는 넓은 흡광도를 가진 잘 알려진 생체 고분자다. 따라서 본 연구는 금 및 멜라닌 (Gold-melanin) 하이브리드 나노 입자는 폐암의 광열 치료를 위해 개발되었다. 나노 입자의 조합은 개별 비해 높은 광열 특성을 갖는 것으로 입증되었다. 그러나, 광열 치료의 나노 입자 타당성을 평가하기 위해, 암 표적 기능이 필요 하였다. Gold-멜라닌 나노 입자가 성공적으로 선택적으로 암세포를 표적으로하기 위해 표피 성장 인자 (EGF) 및polyethylene glycol (PEG)사이의 복합체로 작용화되었다. Gold-멜라닌@ PEG-EGF 라 불리는 생성 된 나노 입자는 높은 생체 적합성, 혈청에서의 우수한 분산 안정성 및 성공적인 세포 내재화를 나타낸다. 셀룰러 수착 결과로부터, Gold-멜라닌 @ PEG-EGF나노 입자는 성공적 결합 및 A549 세포 내재화 할 수 있었고 세포 독성 효과를 유발하지 않았다. 또한, 나노 입자는 선택적 NIR 조사 하에서 암세포를 파괴 높은 광열 효과를있음을 보여준다. 게다가, Gold-멜라닌 @ PEG-EGF 나노 입자는 EGF 수용체를 표시하는 특정 암 세포에 고유하기 때문 A549 세포에서만 관찰할 수 있으며, EGF 수용체 결여 세포 (MCF-7및 HaCaT)에 결합 할 수 없었다고 하였다. 이러한 결과는 Gold-멜라닌 @ PEG-EGF 나노 입자가 A549 폐암 세포에 생체 특정 둘 다 가능한 광열 화제임을 시사한다. 나는 이 나노 입자는 구체적 대상과 근적외선 광을 이용하여 페 암세포를 파괴 할 수있는 실용적인 광열 에이전트로 큰 약속을 잡고 있다고 생각한다.
