The maintenance effect of hexobarbital on drug-metabolizing enzyme activities were investigated in primary cultures of adult rat hepatocytes. Hexobarbital (lmM) was treated to culture media directly at the time of initial plating. The activities of drug-metabolizing enzymes, such as 7-ethoxycoumarin O-deethylase, aryl hydrocarbon hydroxylase, aminopyrine N-demethylase, and DMN demethylase were measured 24hr after culture. Hexobarbital increased the activities of drug-metabolizing enzymes in hormone-supplemented AB medium compared with WOBA-M medium. To elucidate these differences, the activities of 7-ethoxycoumarin O-deethylase of primary culture different culture conditions were examined. Hydrocortisone in AB medium significantly affected the maintenance effect of hexobarbital. The increase of drug-metabolizing enzyme activity by hexobarbital was proportional to the concentration of hydrocortisone supplemented in culture medium.
본 연구에서는 백쥐 간세포의 일차 배양시 배양후 24 시간 이내에 급격히 감소하는 약물 대사 효소의 활성도를 생체와 유사한 수준으로 유지시켜주기 위한 실험으로 hexobarbital 에 의해 약물 대사 효소가 유지되는 기작을 연구하였다. Hexobarbital 을 배양 초기에 1 mM 농도로 처리하였을 때 홀몬이 첨가되지 않은 WOBA-M2 배지에 비해서 홀몬을 첨가해 준 AB 배지에서 약물 대사 효소계의 활성이 잘 유지되었다. 그리고 첨가하여준 홀몬들 중 glucocorticoid 의 일 종인 hydrocortisone 이 hexobarbital 에 의한 약물대사능 유지에 가장 큰 영향을 미치는 것으로 나타났다. 그 밖에 tocopherol 과 aminolevulinic acid 도 영향을 미친 결과를 얻었다. Hydrocortisone 의 영향은 배지 내의 농도에 비례하여 hexobarbital 의 약물 대사능 유지 효과를 높여 주었다.
