The full length canine gastrin gene was isolated from a genomic library by in situ plaque hybridization, in order to understand and elucidate informations on its expression. Employing a canine gastrin cDNA clone as a hybridization probe, over $5\times10^5$ recombinant phages were screened. This revealed two different clones of 12.5 kb and 14.6 kb genomic DNA fragments that share the same 11.0 kb fragment. The nucleotide sequences of seven regions in the isolated genomic DNA, totally 1100 nucleotides-were revealed. Comparison of the determined genomic sequences with the cDNA sequences revealed an intron-exon junction that follows the GT/AG consensus sequences of mRNA processing. This junction corresponds to the junction of intron 1 and exon 2 of the human gastrin gene. Since the location of this junction is identical to the human gene, it seems that the canine genomic structure reveals extensive homology with human.
